Reduced reproductive success is associated with selective constraint on human genes

Summary: Genome-wide sequencing of human populations has shown significant variation in the intensity of purifying selection among different genes, with certain genes under stronger selective constraint, particularly those associated with Mendelian disorders. Damage-causing genetic variants in these highly constrained genes are linked to reduced reproductive success, primarily due to increased childlessness, with a greater impact on males. This reduction in reproductive success, mediated by genetically associated cognitive and behavioral traits, may account for 20% of purifying selection against heterozygous variants that ablate protein-coding genes, influencing gene evolution over time.
