Glutathione-Depleting Nanomedicines for Synergistic Cancer Therapy

Xiaotong Cheng et al.

Summary: Overexpression of glutathione in cancer cells leads to resistance to anticancer therapies, making GSH depletion a potential target for cancer treatments. By combining with mainstream cancer therapies, GSH-depleting agents can enhance cancer cells responses to cell death stimuli.

Na+ inserted metal-organic framework for rapid therapy of bacteria-infected osteomyelitis through microwave strengthened Fenton reaction and thermal effects

Shubin Wei et al.

Summary: In this study, a rapid and noninvasive therapeutic strategy was developed using Prussian blue as a microwave-responsive material to effectively treat bacteria-induced osteomyelitis. The released Fe2+ and Fe3+ from PB could easily penetrate bacterial membrane by microwave, react with H2O2 and GSH inside bacteria, leading to the final death of the bacteria due to the synergistic action of microwave, microwave thermal effect and Fe2+/Fe3+ induced Fenton reaction/GSH consumption. This study provides novel insight for designing remote and noninvasive antibacterial systems for treating deep bacteria-infected diseases.

Antitumour activity of coordination polymer nanoparticles

Salvio Suarez-Garcia et al.

Summary: Nanoscale coordination polymers (NCPs) have attracted researchers' interest due to their therapeutic properties, excellent drug encapsulation yields, surface functionalization, and potential in drug delivery alternatives in medicine. The ability to fine-tune the properties of these nanoparticles and their significant advances in antitumoural applications, particularly in theranostics, make them a promising nanoplatform for cancer treatment. The review emphasizes the advantages of NCPs, describes examples of NCPs for antitumoural applications, and discusses the challenges and future trends in clinical translation.

Ferroptosis-Strengthened Metabolic and Inflammatory Regulation of Tumor-Associated Macrophages Provokes Potent Tumoricidal Activities

Zhengying Gu et al.

Summary: This study demonstrates that an iron-based metal organic framework nanoparticle and a ferroptosis-inducing agent synergistically induce mitochondrial alternation in TAMs, resulting in a radical metabolic switch from mitochondrial oxidative phosphorylation to glycolysis, which is resistant to anti-inflammatory stimuli challenge. The ferroptosis stress strengthened by the nanoformulation also drives multiple pro-inflammatory signaling pathways, enabling macrophage activation with potent tumoricidal activities. The ferroptosis-strengthened macrophage regulation strategy present in this study paves the way for TAM-centered antitumoral treatment to overcome the limitations of conventional methods.

Reinforcing the Induction of Immunogenic Cell Death Via Artificial Engineered Cascade Bioreactor-Enhanced Chemo-Immunotherapy for Optimizing Cancer Immunotherapy

Kai Sun et al.

Summary: Traditional chemo-immunotherapy can induce T cell immune response through immunogenic cell death (ICD), but the limited ICD hinders lasting antitumor immunotherapeutic efficacy. A novel tadpole-ovoid Au@HMnMSNs nanoreactor is constructed to generate intratumoral hydroxyl radicals for enhanced ICD induction and DC maturation, along with DOX and ASA for recruitment of immune cells and inhibition of immunosuppressive cells.
