Summary: Adoptive T-cell transfer (ACT) is a curative therapeutic option for melanoma and hematological cancer patients, and improving the post-infusion performance of transferred T cells can enhance treatment efficacy. In vivo priming and expansion of previously non-activated T cells can boost the cytotoxicity of activated T cells. Flexible delivery vehicles of tumor-associated antigens (TAAs) and a TLR7/8 agonist can effectively enhance T-cell expansion and anti-tumor reactivity.

Summary: The study showed the preliminary feasibility and safety of CRISPR-engineered CAR-T cells with PD-1 disruption in a dose-escalation study with 15 patients, demonstrating no dose-limiting toxicity or unexpected adverse events. Furthermore, it indicated the importance of natural TCR in the persistence of CAR-T cells when treating solid tumors, as TCR-positive CAR-T cells were predominantly detected in effusion or peripheral blood after infusion.
