Bioactive hydrogel microcapsules for guiding stem cell fate decisions by release and reloading of growth factors

The nano-coating on iPSCs allows them to retain their properties such as adhesion capability, proliferation, and pluripotency during aggregation, without inducing lineage-specific differentiation but increasing E-cadherin expression to promote EB development. This provides a foundation for future production of sophisticated patient-specific multicellular aggregates through modification of living cell membranes.

Microfluidic Fabrication of Core-Shell Microcapsules carrying Human Pluripotent Stem Cell Spheroids

Summary: This paper describes a strategy for the robust and reproducible formation of hPSC spheroids using a co-axial flow focusing device. By combining core and shell streams wrapped in hydrogel, microcapsules with an aqueous core were formed, allowing rapid assembly of single cells into aggregates and spheroids. The encapsulation technology yielded high viability and efficient spheroid formation within 48 hours, making it well-suited for hPSC encapsulation and spheroid formation.
