Oriented nanofibrous P(MMD-co-LA)/Deferoxamine nerve scaffold facilitates peripheral nerve regeneration by regulating macrophage phenotype and revascularization

Summary: This study explores the effect of an oriented nanofiber/Deferoxamine (DFO) scaffold in the construction of a favorable regenerative microenvironment for nerve regeneration. In vitro experiments showed that the scaffold sustained release of DFO, promoting the migration and tube formation of endothelial cells and the expression of angiogenesis-related genes. The physical cues provided by the nanofibers regulated macrophage polarization and reduced the expression of inflammatory factors. In vivo results demonstrated reduced inflammation and enhanced nerve regeneration with the use of the oriented nanofibrous scaffold and in-situ release of DFO.
