Efficient target cleavage by Type V Cas12a effectors programmed with split CRISPR RNA

Regina Shebanova et al.

Summary: This study demonstrates that the removal of parts of the CRISPR RNA has minimal effect on the DNA cleavage activity of Cas12a nucleases in vitro. Split CRISPR RNAs can efficiently and specifically catalyze DNA cleavage by Cas12a nucleases in vitro and in human cell extracts. These findings provide insights into the mechanisms of target recognition and cleavage and have implications for the development of diagnostic tests based on Cas12a nucleases.
