Druggable Allosteric Sites in β-Propeller Lectins

A druggable pocket in a beta-propeller lectin from Burkholderia ambifaria has been identified as a potential target for allosteric inhibitors. Future drug-discovery efforts focusing on small molecule inhibitors could benefit from targeting allosteric sites in lectins to combat antibiotic-resistant pathogens.

Non-Carbohydrate Glycomimetics as Inhibitors of Calcium(II)-Binding Lectins

A catechol was identified as an experimentally confirmed hit, acting as a millimolar LecA inhibitor. The first co-crystal structure of a non-carbohydrate inhibitor in complex with a bacterial lectin demonstrated how the catechol mimics the binding of natural glycosides with LecA, indicating a fundamentally new class of glycomimetics.
