Mitochondrial redox regulation and myocardial ischemia-reperfusion injury

Chwen-Lih Chen et al.

Summary: Mitochondrial reactive oxygen species (ROS) play a crucial role in disease mechanisms and redox signaling in the cellular system. Various factors regulate the redox status in mitochondria, including ROS, proton motive force (PMF), oxidative post-translational modifications (OPTM), and SOD2. Pathological conditions, such as oxidative stress, increase mitochondrial ROS production and disrupt redox function through oxidative damage and impairment of heme integrity. Conversely, reductive stress induced by enhanced SOD2 function normalizes ROS overproduction and redox dysfunction. Investigating the regulation of PMF and ROS production by HCCS, biogenesis of c-type cytochrome, and OPTM in mitochondria will contribute to our understanding of redox signal transduction and provide potential therapeutic targets for cardiovascular diseases associated with oxidative stress.

Mitophagy Regulation Following Myocardial Infarction

Annie Turkieh et al.

Summary: Mitophagy plays a crucial role in maintaining cardiac homeostasis by selectively eliminating dysfunctional mitochondria. It is mainly regulated by the PINK1/parkin pathway and also by the FUNDC1, BNIP3, and BNIP3L/NIX pathways. Dysregulated mitophagy is associated with cardiac dysfunction, while moderate mitophagy has a cardioprotective effect.

Interplay of Oxidative Stress and Necrosis-like Cell Death in Cardiac Ischemia/Reperfusion Injury: A Focus on Necroptosis

Adriana Adameova et al.

Summary: This review highlights the importance of regulated necrosis-like cell death program in cardiac ischemic injury. Key molecules of the necroptosis signaling pathway have been found to protect the heart by reducing infarct size and improving myocardial dysfunction. The non-canonical pathway is also associated with ischemic heart injury. Oxidative stress may be involved in the mechanisms of this cell death program.

Intersection of autophagy regulation and circadian rhythms in the heart

Inna Rabinovich-Nikitin et al.

Summary: Autophagy is an important cellular mechanism for removing damaged or dysfunctional cellular components. It plays a crucial role in the cardiovascular system and contributes to normal cardiac function. The activation of autophagy during cardiac stress can be adaptive or maladaptive. This review highlights the molecular mechanisms and signaling pathways underlying autophagy in response to cardiac stress, as well as therapeutic approaches to modulate autophagy. The intersection between autophagy and circadian regulation in the heart is also discussed.

Oxidation of Protein Kinase A Regulatory Subunit PKARIα Protects Against Myocardial Ischemia-Reperfusion Injury by Inhibiting Lysosomal-Triggered Calcium Release

Jillian N. Simon et al.

Summary: The study revealed that PKARI alpha disulfide formation is significantly increased in response to I/R in both humans and mice, leading to enhanced AKAP binding and preferential localization to the lysosome. This redox-dependent regulation of lysosomal channels by PKARI alpha plays a critical role in protecting the heart from extensive injury and could serve as a novel target for cardioprotective therapeutics.

Summary: Ischemic preconditioning and postconditioning cardioprotective strategies target mitochondria to reduce I/R injury by activating protective signaling pathways and MQC processes, maintaining normal mitochondrial function and ensuring cardiomyocyte survival. MQC includes mitochondrial biogenesis, fission, fusion, mitophagy, and mitochondrial-controlled cell death. Modulation of MQC mechanisms and recognition of mitochondrial targets could offer a potential therapeutic approach for I/R-induced mitochondrial dysfunction.
