Summary: This study, the largest genome-wide association study for eye color to date, identified 124 independent associations from 61 genomic regions. In addition to genes related to melanin pigmentation, genes related to iris morphology and structure were also found. The genetic complexity of human eye color was shown to exceed previous knowledge and expectations.

Summary: A GWAS study of human hair color in a Canadian population identified candidate causal variants associated with blonde, red, and brown hair color. Colocalization of GWAS hits with expression and methylation QTLs of cultured melanocytes was observed. Additionally, TWAS identified the expression of EDNRB and CDK10 as significantly associated with hair color.

Cell-type-specific meQTLs extend melanoma GWAS annotation beyond eQTLs and inform melanocyte gene-regulatory mechanisms

Summary: The study established an meQTL dataset using primary melanocytes and identified significant cis-meQTLs, which along with other QTLs helped identify candidate susceptibility genes at melanoma GWAS loci. Additionally, the comparison between melanocyte meQTLs and malignant melanoma meQTLs revealed commonalities and differences, with insights into potential regulatory networks mediated by transcription factor IRF4. This study highlights the importance of cell-type-specific meQTLs in understanding disease susceptibility and molecular mechanisms.
