Repurposing pantoprazole and haloperidol as efflux pump inhibitors in azole resistant clinical Candida albicans and non-albicans isolates

Amira M. El-Ganiny et al.

Summary: Candida species play a major role in nosocomial infections and have become increasingly resistant to antifungals, particularly azoles. This study investigated the mechanisms of azole resistance in clinical Candida isolates and explored the potential synergistic effect of haloperidol and pantoprazole in overcoming fluconazole resistance. The majority of resistant isolates formed biofilm and overexpressed efflux pumps. Pantoprazole and haloperidol showed promising efficacy as efflux pump inhibitors against fluconazole-resistant Candida species.

Candida albicans-The Virulence Factors and Clinical Manifestations of Infection

Jasminka Talapko et al.

Summary: Candida albicans is a common commensal fungus in the human body, which can cause various infections when the balance with the host immune system is disrupted. Factors such as dysbiosis of microbiota, immune dysfunction, and mucosal barrier damage contribute to the transition of C. albicans from a normal flora to a pathogenic state. The presence of C. albicans in the bloodstream can lead to invasive infections.

Summary: The study improved the efficacy of anthocyanins in lipophilic systems by esterifying cyanidin-3-O-glucoside with fatty acids. The derivatives showed lower antioxidant and enzymatic inhibitory activities compared to the precursor, but exhibited a significant biphasic effect in antibacterial activity, particularly against Gram-positive bacteria. Cy3glc-C10 was the most effective compound in terms of antimicrobial activity, prompting further exploration of different derivatives with various features.

Engy Elekhnawy et al.

Summary: The study evaluated the effect of exposure of Proteus mirabilis to sublethal concentrations of Triclosan, finding that most adapted isolates exhibited increased antibiotic resistance, lower membrane integrity, and changes in membrane characteristics and efflux activity. These results suggest that the extensive usage of Triclosan at sublethal concentrations could contribute to the emergence of antibiotic resistance in P. mirabilis clinical isolates.
