Exosomal miRNAs from Prostate Cancer Impair Osteoblast Function in Mice

Giulia Furesi et al.

Summary: Prostate cancer communicates with bone progenitor cells through extracellular vesicles, promoting vitality and proliferation of the progenitor cells while decreasing mineral deposition. Additionally, small RNAs released by prostate cancer cells may play a critical role in suppressing the activity of bone progenitor cells. These findings reveal an important communication mechanism between prostate cancer and bone.

Host lipidome and tuberculosis treatment failure

Rupak Shivakoti et al.

Summary: This study investigated the association between host lipids and treatment failure in tuberculosis (TB) using unbiased lipidomics. The results showed that treatment failure was associated with lower levels of cholesteryl esters and oxylipin, and higher levels of ceramides and triglycerides at the baseline. A lipid signature with predictive accuracy for TB treatment failure was also identified.

Sphingomyelin Biosynthesis Is Essential for Phagocytic Signaling during Mycobacterium tuberculosis Host Cell Entry

Patrick Niekamp et al.

Summary: Phagocytosis by alveolar macrophages is the initial step in Mycobacterium tuberculosis (Mtb) infection. The process relies on an intact sphingolipid biosynthetic pathway, where disruption leads to reduced pathogen uptake. This pathway is crucial for the segregation of regulatory molecules and downstream activation of signaling molecules, highlighting a potential target for antimycobacterial therapeutics.

Nutritional markers and proteome in patients undergoing treatment for pulmonary tuberculosis differ by geographic region

Leah G. Jarsberg et al.

Summary: The study compared the serum proteome of TB patients from African and non-African regions undergoing treatment, finding significant differences in the expression of nutrition and lipid pathway proteins which may be associated with differential treatment responses. Future TB clinical trials should consider collecting expanded measures of nutritional status to further evaluate their relationship with treatment outcomes.

Emerging Role of Exosomes in Tuberculosis: From Immunity Regulations to Vaccine and Immunotherapy

Yin-Fu Sun et al.

Summary: Exosomes are important signal transport vectors for cell-cell communication and immune modulation, playing a critical role in anti-MTB immune response. Infected-cell-derived exosomes trigger different immune responses during tuberculosis infection, highlighting their potential in developing new vaccination and treatment strategies for TB. Exosomes, as a novel kind of delivery system, show promise in the field of novel anti-MTB vaccines and therapies.

Exosomes in serum-free cultures of THP-1 macrophages infected with Mycobacterium tuberculosis

Fantahun Biadglegne et al.

Summary: This study characterized exosomes released from THP-1 macrophages cultured in serum-free, ultra-centrifuged medium upon Mycobacterium tuberculosis infection. The results showed that the serum-free medium supported cell growth and allowed for the successful isolation of exosomes expressing CD9, CD63, and CD81 markers, without the presence of Mtb proteins. This suggests that the use of serum-free medium can offer advantages for exosome research and improve extracellular vesicle studies.
