Targeting ER protein TXNDC5 in hepatic stellate cell mitigates liver fibrosis by repressing non-canonical TGFβ signalling

Chen-Ting Hung et al.

Summary: The study identified TXNDC5 as a critical mediator of liver fibrosis, showing that its upregulation is associated with the activation of HSCs and the production of excessive extracellular matrix. Targeting TXNDC5 could potentially be a novel therapeutic strategy to ameliorate liver fibrosis.

Inhibition of TXNDC5 attenuates lipopolysaccharide-induced septic shock by altering inflammatory responses

Yanping Zeng et al.

Summary: TXNDC5 is highly expressed in patients with septic shock as well as in mice with LPS-induced sepsis and mouse macrophages stimulated with LPS. Depletion of Txndc5 reduces inflammatory cytokine production and affects the recruitment of immune cells in LPS-challenged mice, ultimately alleviating sepsis by inhibiting the NF-kappa B signaling pathway. This suggests that targeting TXNDC5 may be a potential therapeutic approach for sepsis.

Hepatic galectin-3 is associated with lipid droplet area in non-alcoholic steatohepatitis in a new swine model

Luis V. Herrera-Marcos et al.

Summary: This study describes a new experimental swine model for initial and reversible non-alcoholic fatty liver disease (NAFLD). Gene expression analysis revealed several genes associated with steatosis and steatohepatitis. The results suggest that LGALS3 may play an important role in the transition from NAFLD to non-alcoholic steatohepatitis (NASH).

Targeting mechanosensitive endothelial TXNDC5 to stabilize eNOS and reduce atherosclerosis in vivo

Chih-Fan Yeh et al.

Summary: This study provides experimental evidence of the role of flow-induced endothelial TXNDC5 in atherosclerosis development. It shows that TXNDC5 is increased in atherosclerotic lesions and induced in endothelium subjected to disturbed flow. The study also demonstrates that targeting TXNDC5 can reduce degradation of heat shock protein 90 and increase eNOS protein expression, leading to reduced atherosclerosis.
