The Impact of Systemic Inflammation on Alzheimer's Disease Pathology

Junhua Xie et al.

Summary: Alzheimer's disease is an age-related neurodegenerative disorder with increasing prevalence. Current treatments are limited to symptomatic medication, and there is a need for a better understanding of the underlying mechanisms to develop more rational therapies. Peripheral inflammation has been identified as a detrimental factor in AD, and this review provides an overview of the link between peripheral inflammation and AD initiation and progression, contributing to a broader understanding of the disease pathology.

Risk of Dementia or Cognitive Impairment in Sepsis Survivals: A Systematic Review and Meta-Analysis

Summary: Recent research suggests that sepsis survivors may be at higher risk for developing dementia, but the association with cognitive impairment is less clear. Severe sepsis appears to be more strongly associated with increased risk of dementia or cognitive impairment.

Neuroinflammation and microglial activation in Alzheimer disease: where do we go from here?

Fangda Leng et al.

Summary: Microglial activation and neuroinflammation play important roles in Alzheimer's disease, affecting disease progression and pathologies. Current research focuses on the interplay between neuroinflammation and amyloid and tau pathologies, with a specific interest in modulating microglia as a therapeutic strategy for AD.

Summary: This study found that in individuals with higher Aβ and tau burden, the association between small vessel pathologies and cortical microinfarcts was stronger, suggesting a potential link between neurodegenerative and vascular mechanisms.

Low-grade peripheral inflammation affects brain pathology in the AppNL-G-Fmouse model of Alzheimer's disease

Junhua Xie et al.

Summary: Alzheimer's disease is characterized by accumulation of amyloid beta and neurofibrillary tangles, with neuroinflammation playing a key role in its progression. Low-grade peripheral inflammation affects microglia characteristics, blood-cerebrospinal fluid barrier integrity, immune cell infiltration, and A beta deposition in the brain in the APP knock-in mouse model, potentially through mechanisms like A beta efflux and neuronal dysfunction. These findings suggest modulating peripheral inflammation could be a therapeutic strategy for AD.

Plasma markers predict changes in amyloid, tau, atrophy and cognition in non-demented subjects

Joana B. Pereira et al.

Summary: Plasma biomarkers, particularly Aβ 42/40 and phosphorylated-tau217, can predict future Alzheimer's disease pathology in non-demented individuals, making them potential prognostic markers for clinical practice, research, and drug development.

Blood-based high sensitivity measurements of beta-amyloid and phosphorylated tau as biomarkers of Alzheimer's disease: a focused review on recent advances

Joyce R. Chong et al.

Summary: Recent research efforts have focused on discovering and developing clinically useful biomarkers for AD and related dementias. While advancements in blood-based biomarkers hold promise for diagnosis, prognosis, and clinical trial outcome assessments, further studies are needed to establish A beta and P-tau as blood-based biomarkers for AD.

Don't Phos Over Tau: recent developments in clinical biomarkers and therapies targeting tau phosphorylation in Alzheimer's disease and other tauopathies

Yuxing Xia et al.

Summary: Phosphorylation is a common post-translational modification in tau protein isolated from Alzheimer's disease patients, with therapeutic approaches targeting phosphorylated tau showing promise in slowing hyperphosphorylation and aggregation. The identification and monitoring of phosphorylated tau could serve as disease-specific biomarkers, aiding in clinical diagnosis and progression monitoring.

Oxidative Stress and Beta Amyloid in Alzheimer's Disease. Which Comes First: The Chicken or the Egg?

Elena Tamagno et al.

Summary: The pathogenesis of Alzheimer's disease involves the accumulation of beta amyloid and vulnerability of the brain to oxidative stress, which are linked to each other. It is difficult to determine which comes first, Aβ or oxidative stress. Evidence suggests that oxidative stress occurs early in the development of Alzheimer's disease and plays a crucial role in the manifestation of clinical and pathological symptoms.
