Identification of proteins associated with development of metastasis from cutaneous squamous cell carcinomas (cSCCs) via proteomic analysis of primary cSCCs

A. Shapanis et al.

Summary: The study identified that increased expression of proteins ANXA5 and DDOST is significantly associated with poorer clinical outcomes in cSCC, and can be used to predict the metastasis and survival of patients. The prediction model using these proteins showed higher accuracy, sensitivity, and specificity than current cSCC staging systems in clinical use.

Clinical relevance of blood-based ctDNA analysis: mutation detection and beyond

Laura Keller et al.

Summary: Cell-free DNA derived from tumors can be detected in the plasma of cancer patients, and significant progress has been made in the detection of mutations in circulating tumor DNA. In addition to mutation detection, cfDNA can be used to analyze other features of DNA, providing new avenues for liquid biopsy analysis.

Summary: In patients with HPV16-related HNSCC, posttreatment ctHPV16DNA has a higher value in predicting treatment failure compared to PET-CT. Patients with detectable posttreatment ctHPV16DNA have a higher risk of treatment failure regardless of metabolic response.

The effect of age on the acquisition and selection of cancer driver mutations in sun-exposed normal skin

B. Hernando et al.

Summary: This study reveals that UV-related somatic mutations accumulate exponentially with age in normal skin, matching skin cancer incidence. The increase in mutation burden is influenced by an individual's skin type, and somatic mutations tend to accumulate and clonally expand in specific cancer genes as individuals age.

The Genomic Landscape of Actinic Keratosis

Jason Thomson et al.

Summary: This study conducted the largest AK whole exome sequencing study to date and found significant similarities between AK and cSCC in terms of genetic mutations, copy number alterations, driver gene mutations, and mutational signatures. The research identified 44 significantly mutated AK driver genes and suggested that dysregulated TGFf3 signaling may play an important role in AK-cSCC progression.

The Blood Microenvironment Influences the Molecular Phenotypes of Circulating Tumor Cells in Head and Neck Squamous Cell Carcinoma

Hiroe Tada et al.

Summary: The study found that CTCs acquire a survival advantage through phenotypic alterations in the hostile blood environment and evade circulatory surveillance. Expression of VIM or ZEB2 in CTCs was strongly correlated with CD44 or ALDH1, PIK3CA, CD44, ALDH1A1, and PDCD1LG2 expressions. Additionally, expression of these genes in CTCs was correlated with systemic inflammatory markers related to lymphocytes and/or albumin.

Serum matrix metalloproteinase-13 as a diagnostic biomarker for cutaneous squamous cell carcinoma

Hui Wang et al.

Summary: Serum MMP-13 levels are significantly higher in patients with cSCC compared to healthy controls, particularly in those with invasive cSCC. Surgical removal of cSCC results in a significant decrease in serum MMP-13 levels. Serum MMP-13 can predict the invasiveness of cSCC and lymph node involvement with high sensitivity and specificity.

Integrated tissue proteome and metabolome reveal key elements and regulatory pathways in cutaneous squamous cell carcinoma

Wen Chen et al.

Summary: This study comprehensively revealed the molecular characteristics of cutaneous squamous cell carcinoma (CSCC) by performing integrated tissue proteomic and metabolomic analyses, identifying significantly distinct profiles and enriched pathways associated with CSCC, including DNA damage responses. Protein digestion & absorption and platelet activation were both enriched in the proteome and metabolome, suggesting significant roles in CSCC development. Additionally, a four-metabolite panel was developed to distinguish CSCC with high accuracy, providing potential diagnostic biomarkers and extending knowledge on the pathogenesis of CSCC.

The landscape of driver mutations in cutaneous squamous cell carcinoma

Darwin Chang et al.

Summary: Cutaneous squamous cell carcinoma originates from keratinocytes in the skin and is characterized by high mutation burden primarily caused by UV-induced DNA damage. Through meta-analysis of sequencing data, multiple driver genes were identified, offering potential therapeutic targets and insights into the biology of this type of skin cancer.
