A Two-Stage Model for Predicting Mild Cognitive Impairment to Alzheimer's Disease Conversion

Peixin Lu et al.

Summary: This study proposes a two-stage model combining transfer learning and contrastive learning for early Alzheimer's disease (AD) diagnosis based on MRI. The model achieves high accuracy even with limited sample numbers and enhances interpretability using 3D Grad-CAM.

Prediction of Cognitive Decline for Enrichment of Alzheimer's Disease Clinical Trials

A. Tam et al.

Summary: A key issue in Alzheimer's disease clinical trial failures is poor participant selection. This study developed machine learning models to predict cognitive trajectories in individuals with early Alzheimer's disease and presymptomatic individuals, aiming to recruit enriched cohorts of predicted decliners and improve trial quality and therapeutic development in Alzheimer's disease.

Diagnostic accuracy of the Clinical Dementia Rating Scale for detecting mild cognitive impairment and dementia: A bivariate meta-analysis

Hui-Chuan Huang et al.

Summary: The CDR shows good diagnostic accuracy for MCI and dementia in older populations. It has high sensitivity and specificity for diagnosing MCI, with better specificity for dementia in CDR-GS compared to CDR-SB. Various factors such as older age, high education level, high prevalence of MCI or dementia, being in a developing country, and lack of informant observations may affect the accuracy of CDR in diagnosis.

Cognitive and MRI trajectories for prediction of Alzheimer's disease

Samaneh A. Mofrad et al.

Summary: The concept of Mild Cognitive Impairment (MCI) is used to describe early stages of Alzheimer's disease, and the study showed that predictions between normal function and MCI improved significantly when MRI features were included. The distinction between stable MCI and conversion to AD had high accuracy regardless of MRI features.

Machine Learning and Novel Biomarkers for the Diagnosis of Alzheimer's Disease

Chun-Hung Chang et al.

Summary: This study investigated the application of machine learning and novel biomarkers in the diagnosis of Alzheimer's disease (AD). In addition to traditional Aβ and tau-related biomarkers, biomarkers related to neuronal injury, synaptic dysfunction, and neuroinflammation were explored. Machine learning combined with novel biomarkers and multiple variables may improve the sensitivity and specificity in diagnosing AD.

The Measurement of Everyday Cognition (ECog) Revisions and Updates

Sarah T. Farias et al.

Summary: The Everyday Cognition scale (ECog), developed in 2008, has been updated to ensure relevancy to contemporary older adults from diverse backgrounds. Through evaluation and modification of items with input from culturally diverse participants, the revised ECog (ECog-II) now includes 41 items and covers contemporary activities such as the use of technology. This updated version will continue to be a useful tool for measuring cognitively relevant everyday abilities in clinical settings and intervention clinical trials.

Delirium in critical illness: clinical manifestations, outcomes, and management

Joanna L. Stollings et al.

Summary: Delirium is a common manifestation of brain dysfunction in critically ill patients, with its duration predicting various negative outcomes. The absence of validated tools may lead to misdiagnosis, while ICU bedside instruments can provide valuable information. Screening for predisposing and precipitating factors and implementing strategies like the ABCDEF bundle can improve patient outcomes.

Evelyn Fokuoh et al.

Summary: This study investigated the longitudinal effect of the APOE-ε4 genotype on logical memory declines in late-onset Alzheimer's disease, showing significant differences in LDELTOTAL scores among AD, CN, and MCI groups. Individuals with 1 or 2 APOE-ε4 alleles had significantly lower LDELTOTAL scores, and APOE-ε4 alleles interacted with gender and follow-up visits.

Delirium and long term cognition in critically ill patients

M. Elizabeth Wilcox et al.

Summary: Delirium, a common acute brain dysfunction in critically ill patients, lacks clear understanding of its pathophysiology and effective treatments. Long duration of delirium is associated with poor long term cognition, and there are differences in cognitive impairment between ICU survivor populations without clear identification of patients at higher risk of long term cognitive impairment.

Neuroinflammation and Alzheimer's Disease: A Machine Learning Approach to CSF Proteomics

Lorenzo Gaetani et al.

Summary: In this study, novel protein biomarkers reflecting neuroinflammation in Alzheimer's disease were identified using a high-throughput multiplex CSF analysis coupled with machine-learning. These biomarkers, including SIRT2, HGF, MMP-10, and CXCL5, showed high discriminatory performance in distinguishing AD patients from controls. The biological processes regulated by these proteins involve astrocyte and microglia activation, modulation of amyloid and tau misfolding, and dysfunction of the blood-brain barrier. Additional studies are needed to confirm these results and validate proximity extension assay as a technique for biomarker discovery in neurological diseases.
