Glioblastoma mutations alter EGFR dimer structure to prevent ligand bias

Chun Hu et al.

Summary: The epidermal growth factor receptor (EGFR) frequently mutates in human cancer, and is an important therapeutic target. However, EGFR inhibitors have limited effectiveness in glioblastoma multiforme (GBM) due to exclusive mutations in the extracellular region. This study reveals that GBM mutations impair EGFR's ability to distinguish between activating ligands, which may have implications for therapeutic targeting.

A Novel 16-Genes Signature Scoring System as Prognostic Model to Evaluate Survival Risk in Patients with Glioblastoma

Zunpeng Yu et al.

Summary: This study aimed to identify prognostic survival-related genes for glioblastoma (GBM) and construct a survival risk prediction model. By analyzing gene expression data and clinical information, 99 genes associated with overall survival were identified and a model with 16 genes was established. The model showed superior prediction ability compared to existing signatures and the genes could potentially serve as transcriptional biomarkers, aiding in personalized treatment for GBM patients.

Prognostic value and immune relevancy of a combined autophagy-, apoptosis- and necrosis-related gene signature in glioblastoma

Ying Bi et al.

Summary: In this study, we developed a 16-gene cell death index (CDI) based on genes related to autophagy, apoptosis, and necrosis and found that it can be used as a prognostic predictor for glioblastoma patients. The CDI score divided patients into high risk and low risk groups, with the low risk group showing significantly longer overall survival and progression-free survival. The gene signature also demonstrated excellent performance in both training and validation groups. Immune cell analysis revealed differences in the immune microenvironment between the two prognostic groups, suggesting potential targets for immunotherapy.

An absolute human stemness index associated with oncogenic dedifferentiation

Hailong Zheng et al.

Summary: The study introduced an absolute REO-based stemness index to evaluate the stemness, showing higher correlation with culture time compared to previous methods. The stemness index demonstrated significant differences between cancer and normal tissues, as well as the ability to reveal intratumor heterogeneity at the stemness level. Higher stemness index was associated with poorer prognosis and greater oncogenic dedifferentiation, indicating its potential therapeutic and diagnostic implications.

Ferroptosis-related gene AKR1C1 predicts the prognosis of non-small cell lung cancer

Fangfang Huang et al.

Summary: The study identified the potential prognostic value of ferroptosis-related gene AKR1C1 in non-small cell lung cancer (NSCLC). Elevated expression of AKR1C1 was associated with low levels of ferroptosis and silencing AKR1C1 inhibited cell proliferation and migration while promoting ferroptosis occurrence in NSCLC. This suggests AKR1C1 could serve as a prognostic predictor in NSCLC.

Broadening horizons: the role of ferroptosis in cancer

Xin Chen et al.

Summary: Ferroptosis is an iron-dependent form of regulated cell death driven by excessive lipid peroxidation, and can be induced by various agents to suppress tumor growth while also potentially triggering inflammation-associated immunosuppression. The extent of ferroptosis's impact on tumor biology and its interactions with other signaling pathways are still under investigation.

The Role of Plasmacytoid Dendritic Cells in Cancers

Binhui Zhou et al.

Summary: Plasmacytoid dendritic cells (pDCs) are a special subtype of dendritic cells that produce massive amounts of type I interferon (IFN-I) and play a pivotal role in antiviral immunity. However, in many types of cancer, tumor-infiltrating pDCs do not respond to Toll-like receptor (TLR) stimulation, leading to reduced IFN-alpha production and promoting tumor growth.
