Adaptive laboratory evolution accelerated glutarate production by Corynebacterium glutamicum

Programming adaptive laboratory evolution of 4-hydroxyisoleucine production driven by a lysine biosensor in Corynebacterium glutamicum

Summary: In this study, programming adaptive laboratory evolution (ALE) was conducted in Corynebacterium glutamicum to promote 4-Hydroxyisoleucine (4-HIL) biosynthesis, resulting in the successful generation of nine mutant strains with increased production. The maximum 4-HIL titer achieved was 152.19 +/- 14.60 mM, surpassing all previously engineered C. glutamicum strains. Genomic analysis revealed approximately 30 genetic mutations in each evolved strain, with only one mutation directly related to the metabolic pathway. Therefore, programming ALE using a biosensor-driven approach can be an effective strategy to enhance 4-HIL production in C. glutamicum.

Evolving a New Efficient Mode of Fructose Utilization for Improved Bioproduction in Corynebacterium glutamicum
