A chromosomal loop anchor mediates bacterial genome organization

Gaurav Dugar et al.

Summary: A study reveals the existence of large nucleoprotein complexes formed by the transcription factor Rok in the bacterium Bacillus subtilis. These complexes can interact with each other over long distances and lead to the formation of chromosomal loops, spatially isolating large sections of DNA.

Molecular basis for the adaptive evolution of environment-sensing by H-NS proteins

Xiaochuan Zhao et al.

Summary: H-NS, a DNA-binding protein in gram-negative bacteria, plays a pleiotropic role in gene regulation by sensing temperature and other environmental factors. Comparison of H-NS orthologs from different bacteria species revealed how the same mechanism was adapted to specific habitats and lifestyles, with environment-sensing characteristics arising from specific intra- or intermolecular electrostatic interactions. This study clarifies the modus operandi for H-NS-mediated environmental sensing and suggests that this mechanism resulted from the exaptation of an ancestral protein feature.

DNA sequence-directed cooperation between nucleoid-associated proteins

Aleksandre Japaridze et al.

Summary: This study investigated the formation of long-range nucleoprotein structures by two major NAPs, FIS and H-NS, binding to DNA molecules with distinct binding site arrangements using atomic force microscopy and solid-state nanopores. The spatial organization of protein binding sites was found to govern the higher-order architecture of the nucleoprotein complexes, highlighting the role of DNA sequence in driving structural differentiation within the bacterial chromosome.
