Sodium alginate microencapsulation of human mesenchymal stromal cells modulates paracrine signaling response and enhances efficacy for treatment of established osteoarthritis

Jay M. McKinney et al.

Summary: The study evaluated the effects of sodium alginate microencapsulation on human MSCs in treating OA, and found that encapsulated MSCs can reduce cartilage degeneration and bone remodeling. In response to IL-1 beta stimulation, encapsulated MSCs showed a targeted secretory response, while nonencapsulated MSCs showed an indiscriminate increase in cytokines.

Injection-Free Delivery of MSC-Derived Extracellular Vesicles for Myocardial Infarction Therapeutics

Junnan Tang et al.

Summary: This study proposes a non-injection method for delivering EVs onto the heart surface to treat MI by spraying a mixture of EVs, GelMA precursors, and photoinitiators. The physically entrapped EVs within the GelMA hydrogel network have an enhanced retention rate and are gradually released through diffusion and/or enzymatic degradation to exert a therapeutic effect in the heart tissue.

Immuno-suppressive hydrogels enhance allogeneic MSC survival after transplantation in the injured brain

Melissa Alvarado-Velez et al.

Summary: The development of immunosuppressive hydrogels for stem cell delivery, such as the agarose-FasL hydrogel in this study, has shown promising results in enhancing the benefits of stem cell therapy for traumatic brain injury. By reducing host cytotoxic CD8(+) T cell populations, increasing the survival of transplanted allogeneic MSCs, and elevating the expression of neurotrophic factors in the injury penumbra, this approach holds potential for improving therapeutic outcomes in TBI cases.

Bioactive decellularized cardiac extracellular matrix-based hydrogel as a sustained-release platform for human adipose tissue-derived stromal cell-secreted factors

Tacia Tavares Aquinas Liguori et al.

Summary: The study explored the potential of using dECM hydrogels to load and release paracrine factors secreted by (mesenchymal) cells for cardiovascular regeneration therapy. Different trophic factors exhibited diverse release kinetics depending on the concentration of CMed in the hydrogel. Higher concentrations of hydrogel resulted in a greater release of inflammation-related cytokines and a reduced release of pro-regenerative TFs. The biologically active factors released by the hydrogel influenced cell behavior, indicating the potential of dECM hydrogels as a platform for cardiovascular regeneration.

Tailoring the proliferation of fibroblast cells by multiresponsive and thermosensitive stem cells composite F127 hydrogel containing folic acid. MgO:ZnO/chitosan hybrid microparticles for skin regeneration

Malihe Rafie et al.

Summary: A novel hydrogel containing folic acid.MgO:ZnO/chitosan hybrid particles was developed for delivering mesenchymal stem cells, showing high viability and proliferation rates. The hydrogel exhibited antibacterial activity and stimulated skin cell behavior, making it a promising scaffold for bacteria-accompanied wound healing. The incorporation of FMZC particles in the hydrogel enhanced its biological effects, with synergistic effects observed due to the release of nanocomposites and paracrine factors.

Summary: A potential treatment for chronic wounds using a hydrogel loaded with secretome from adipose tissue mesenchymal stem cells has been proposed. The biocompatibility and pro-angiogenic activity of the hydrogel-released factors were investigated, showing positive effects on skin-origin cell proliferation and endothelial cell angiogenic properties. Additionally, antimicrobial activity was observed in the secretome, indicating its potential use as an advanced wound-healing dressing in combination with the hydrogel.

Cell-Laden Bioactive Poly(ethylene glycol) Hydrogels for Studying Mesenchymal Stem Cell Behavior in Myocardial Infarct-Stiffness Microenvironments

Christopher B. Sylvester et al.

Summary: Cellular therapy with mesenchymal stem cells shows promise for restoring function after myocardial infarction, but difficulty in studying MSCs interaction with the post-MI scar microenvironment has hindered clinical translation. This study successfully designed an in vitro model to study MSC behavior in the post-MI scar stiffness microenvironment, revealing that a 3D microenvironment hinders key mechanisms by which MSCs could improve cardiac function. Future studies should further explore the post-MI scar microenvironment with other cell types to broaden understanding.
