Metformin improves relevant disease parameters in an autosomal dominant polycystic kidney disease mouse model

Nuria M. Pastor-Soler et al.

Summary: This study tested the effectiveness of metformin as a therapy for ADPKD in a relevant mouse model and found that metformin reduced kidney cysts, improved kidney function, lowered blood pressure and urea nitrogen levels, and reduced inflammation and injury markers. These findings have implications for new ADPKD therapies in patients.

Macroautophagy supports Sonic Hedgehog signaling by promoting Patched1 degradation

Summary: This study found that autophagy participates in the ubiquitination and degradation process of Ptch1, thereby regulating the transduction of the Shh signal. Blocking autophagy affects the transport of Smoothened into primary cilia, resulting in weakened Shh signaling.

Regulation of autophagosome biogenesis by OFD1-mediated selective autophagy

Manuela Morleo et al.

Summary: A negative feedback mechanism limiting autophagosome biogenesis by degrading ATG13 is discovered, with OFD1 acting as an autophagy receptor for ATG13. Mutations in the OFD1 gene may lead to excessive autophagy, implicating dysregulated autophagy in the pathogenesis of renal cystic disease in mammals.

A non-canonical Hedgehog pathway initiates ciliogenesis and autophagy

Tara Akhshi et al.

Summary: The study reveals that Smo induces ciliogenesis through two distinct pathways while also activating autophagy, with the Gα(i)-LGN-NuMA-dynein complex playing a crucial role in the process. Different Smo agonists activate these pathways separately, and only together they can replicate the activity of Hh ligand.

Summary: The study demonstrates the important role of IFT-A's IFT144-IFT122 and IFT-B's IFT88-IFT52 in mediating ciliary transport through their cooperation, facilitating the entry of GPCRs and retrograde trafficking of the IFT machinery from the ciliary tip.

A Model for Primary Cilium Biogenesis by Polarized Epithelial Cells: Role of the Midbody Remnant and Associated Specialized Membranes

Leticia Labat-de-Hoz et al.

Summary: Primary cilia are microtubule-based protrusions surrounded by a ciliary membrane equipped with selected receptors that control important signaling pathways in cell growth and development. This review presents a model of primary ciliogenesis by an alternative route involving the midbody remnant from cytokinesis, which delivers membranes to the centrosome for ciliary membrane assembly. The involvement of the midbody remnant in primary cilium formation and its regulation by the ESCRT machinery are discussed, along with the emerging roles of membrane compartmentalization.

The TBC1D31/praja2 complex controls primary ciliogenesis through PKA-directed OFD1 ubiquitylation

Emanuela Senatore et al.

Summary: A novel complex assembled at centrosomes by TBC1D31, including praja2, PKA, and OFD1, is essential for ciliogenesis. PKA phosphorylates OFD1 at ser735 upon GPCR-cAMP stimulation, promoting OFD1 proteolysis through the praja2-UPS circuitry. Disruption of this control mechanism may lead to human genetic disorders.

Jorg Schweiggert et al.

Summary: The ubiquitin E3 ligase SCFFbxw5 targets MCAK for proteasomal degradation during G(2), which promotes ciliogenesis starting in the preceding cell cycle phase. Loss of Fbxw5 results in increased MCAK levels at basal bodies and impairs ciliogenesis in the following G(1)/G(0).
