Molecular interactions of the CTLA-4 cytoplasmic region with the phosphoinositide 3-kinase SH2 domains

Summary: In this study, the crystal structure of a CTLA-4-derived phosphopeptide in complex with a mutant of cSH2 was determined, revealing the molecular mechanism of signal transduction pathways. The Cys mutation increased the thermal stability of cSH2 but had a slight impact on the CTLA-4 binding affinity. The binding experiments showed that the affinity of CTLA-4 by cSH2 was two orders of magnitude lower than that of CD28, with nSH2 also exhibiting lower binding affinity compared to cSH2. The differences in binding affinity between CD28 and CTLA-4, as well as between nSH2 and cSH2, could be explained by the 3D structures and are closely related to T-cell signaling.

The Association Between CTLA-4, CD80/86, and CD28 Gene Polymorphisms and Rheumatoid Arthritis: An Original Study and Meta-Analysis

Summary: The study found that the CTLA-4 rs231775 gene polymorphism decreased the risk of RA, while the CTLA-4 rs16840252 and CD86 rs17281995 gene polymorphisms were not associated with RA susceptibility. Stratification analyses revealed significant associations for CTLA-4 rs231775 and rs16840252 gene polymorphisms in RF-positive and RF-negative groups. The meta-analysis of CTLA-4 gene polymorphisms indicated that CTLA-4 rs231775 and rs3087243 gene polymorphisms decreased the risk of RA. Additional clinical cases should be investigated in further studies to confirm these findings.
