Dendritic Cells Revisited

Mar Cabeza-Cabrerizo et al.

Summary: Dendritic cells play a crucial role in shaping adaptive and innate immunity by integrating environmental information. Conventional DCs, a discrete cell lineage, are highlighted in this review for their key features and impact on other leukocytes.

The inhibitory receptor TIM-3 limits activation of the cGAS-STING pathway in intra-tumoral dendritic cells by suppressing extracellular DNA uptake

Alvaro de Mingo Pulido et al.

Summary: Blockade of the inhibitory receptor TIM-3 has shown efficacy in clinical trials of cancer immunotherapy, specifically in enhancing antitumor immunity in mammary carcinomas by increasing CXCL9 expression by cDC1 cells. This increased expression required type I interferons and extracellular DNA, with DNA uptake and efficacy of TIM-3 blockade being dependent on HMGB1 and galectin-9-induced cell surface clustering of TIM-3, suggesting a potential mechanism for TIM-3 immunotherapy.

Tim-3 adaptor protein Bat3 is a molecular checkpoint of T cell terminal differentiation and exhaustion

Chen Zhu et al.

Summary: T cell exhaustion is associated with poor prognosis in persistent viral infection and cancer, but has a favorable correlation with long-term clinical outcomes in the context of autoimmunity. The adaptor molecule Bat3 acts as a molecular checkpoint of T cell exhaustion, suppressing autoreactive T cell-mediated neuroinflammation by inhibiting mTORC2 and Akt function. Bat3 deficiency leads to increased Akt activity and T cell dysfunction under proinflammatory autoimmune conditions.

TIM-3 restrains anti-tumour immunity by regulating inflammasome activation

Karen O. Dixon et al.

Summary: Research demonstrates the crucial role of TIM-3 in regulating dendritic cell (DC) function, with TIM-3 deletion promoting strong anti-tumour immunity by preventing DCs from expressing a regulatory program and facilitating the maintenance of CD8(+) effector and stem-like T cells. The absence of TIM-3 in DCs leads to increased accumulation of reactive oxygen species, resulting in NLRP3 inflammasome activation, and inhibition of inflammasome activation or downstream effector cytokines IL-1 beta and IL-18 abrogates the protective anti-tumour immunity observed with TIM-3 deletion in DCs.
