Hepatocyte pyroptosis and release of inflammasome particles induce stellate cell activation and liver fibrosis

Summary: The study found that hepatocyte pyroptosis, caused by NLRP3 inflammasome activation, leads to the release of inflammasome proteins and exacerbates fibrogenesis. Inhibition of caspase-1 and gasdermin D activation can prevent pyroptosis. The released inflammasomes are taken up by hepatic stellate cells, promoting inflammation and fibrosis. These findings suggest a novel mechanism for liver injury and potential therapeutic targets for chronic liver diseases.

Understanding the morphological characteristics and mechanisms of neovascularization in the progression from liver fibrosis to cirrhosis and HCC, as well as the efficacy of anti-angiogenesis therapy at different stages, is crucial for improving treatment outcomes for patients with liver diseases.
