In Vitro Comparative Study of Solid Lipid and PLGA Nanoparticles Designed to Facilitate Nose-to-Brain Delivery of Insulin

Summary: The alteration in brain insulin metabolism is considered a pathophysiological factor in Alzheimer's disease. Intranasal administration of insulin via solid lipid nanoparticles and poly(lactic-co-glycolic acid) nanoparticles enhances drug permeability to the brain, with chitosan-coating further improving delivery. These nanoparticles show promising physicochemical properties, increased mucoadhesion, nasal diffusion, and drug release rate compared to native insulin, with chitosan-coated SLNs showing superior performance.

Summary: In this study, Rufinamide-loaded chitosan nanoparticles were formulated and suspended in a thermoresponsive polymer-tamarind seed xyloglucan solution to create a nasal in situ gel for direct nose to brain drug delivery. The optimized nanoparticle formulation had favorable characteristics, and the in situ gelling formulation showed promising results in terms of direct transport efficiency and percentage.
