Contextual dependencies expand the re-usability of genetic inverters

Huseyin Tas et al.

Summary: The implementation of Boolean logic circuits in cells is an active area in synthetic biology, with 20 genetic NOT gates characterized in seven different bacterial contexts to understand interoperability and portability. The study shows that the host context plays a crucial role in determining the performance of genetic circuits.

Standardization of inducer-activated broad host range expression modules: debugging and refactoring an alkane-responsive AlkS/PalkB device

Alejandro Arce-Rodriguez et al.

Summary: The research proposed a compositional standard for debugging and reassembling all regulatory parts involved in genetic expression devices to increase the predictability of gene expression in various bacteria. Despite extensive editing of DNA sequences, the refactored module maintained the original functioning parameters when transferred to different bacterial hosts. The application of this standard to other promoter/regulator pairs could enable more complex genetic programming in non-model bacteria.
