Extracellular vesicles in anti-tumor immunity

Elisabetta Vergani et al.

Summary: The impact of extracellular vesicles (EVs) on anti-tumor immune responses is still being explored. The small size and numerous subtypes of EVs, as well as the difficulty in determining their origin, pose challenges for investigation. While the study of cancer EVs has been facilitated by the existence of tumor cell lines, understanding the phenotypes and functions of immune cell EVs in this context is more complex. The emergence of immunotherapy with immune checkpoint inhibitors has increased the need to understand the role of EVs in immune activation and response, particularly in unraveling and preventing resistance in patients.

The exosomes derived from CAR-T cell efficiently target mesothelin and reduce triple-negative breast cancer growth

Pengxiang Yang et al.

Summary: Exosomes derived from CAR-T cells targeting MSLN showed potential in inhibiting the growth of MSLN-positive triple-negative breast cancer cells, with characteristics similar to parental T cells. The expression of effector molecules may contribute to tumor killing, and in vivo studies demonstrated a highly effective tumor inhibition rate with no obvious side effects. The use of CAR-T cell exosomes has promising therapeutic potential against MSLN-expressing TNBC.

Intra- and Extracellular Effector Vesicles From Human T And NK Cells: Same-Same, but Different?

Marcus Lettau et al.

Summary: CTL and NK cells use overlapping effector mechanisms to eliminate target cells, with effector proteins stored in lysosome-related effector vesicles. These vesicles not only contain cytolytic effectors, but also other immunomodulatory proteins, suggesting a direct relation between intra- and extracellular vesicles. This indicates a potential role for extracellular vesicles as additional effector vesicles or intercellular communicators in a systemic manner.

Cytoskeletal control of the secretory immune synapse

Tiphaine Douanne et al.

Summary: The immune synapse serves as a crucial site for secretion between immune cells, with mechanisms such as centrosome polarization and rapid actin dynamics playing key roles. Cellular networks controlling T cell persistence in adoptive cell therapy

Jack D. Chan et al.

Summary: The differentiation status of tumor-specific T cells greatly impacts their anti-tumor activity, with less differentiated T cells showing better therapeutic effects due to their enhanced expansion and long-term persistence. Adoptive transfer of T cells with stem-like memory or precursor phenotype is associated with improved therapeutic outcomes in cancer patients. Advances in understanding T cell differentiation at the epigenetic and transcriptional levels have led to the development of novel methods for generating more persistent and functional tumor-specific T cells.
