Drug repositioning against COVID-19: a first line treatment

Nousheen Bibi et al.

Summary: COVID-19 vaccine will help attenuate the disease progression rates, but its distribution may pose challenges and delays. Repurposed drugs against SARS-CoV-2 could be an attractive strategy, especially in poorly-resourced countries. This study conducted virtual screening of broad-spectrum antiviral agents against key proteins of SARS-CoV-2, identifying drugs with high binding affinities to multiple targets.

Olfactory regulation by dopamine and DRD2 receptor in the nose

Hai-Qian Zhou et al.

Summary: Olfactory behavior is crucial for animal survival, and olfactory dysfunction is commonly seen in various diseases. This study reveals that mature olfactory sensory neurons (OSNs) express high levels of dopamine D2 receptor (Drd2), which can modulate odorant recognition response by inhibiting odor-induced signaling. Furthermore, nasal dopamine and DRD2 receptor are identified as potential peripheral targets for olfactory modulation.

LRR-protein RNH1 dampens the inflammasome activation and is associated with COVID-19 severity

Giuseppe Bombaci et al.

Summary: The protein RNH1 has been found to inhibit the activation of inflammasomes, reducing inflammation. Deletion of RNH1 in macrophages leads to increased production of IL-1β and activation of caspase-1. Furthermore, RNH1 protein levels were negatively correlated with disease severity and inflammation in COVID-19 patients.

Is Toll-like receptor 4 involved in the severity of COVID-19 pathology in patients with cardiometabolic comorbidities?

Simone Cristina Soares Brandao et al.

Summary: The severe form of COVID-19 is characterized by an abnormal and exacerbated immune response, especially in patients with obesity, diabetes, hypertension, and atherosclerosis. The virus spike protein interacts strongly with TLR4, leading to a cytokine storm and multiple organ failure in some cases.

COVID-19 and neurocognitive disorders

Elizabeta B. Mukaetova-Ladinska et al.

Summary: The review discusses the neurobiological explanation for neurocognitive disorders caused by the COVID-19 pandemic, focusing on viral-induced inflammation, mechanisms of viral entry into the central nervous system, and altered neurotransmitter systems. The symptoms of mild and major neurocognitive disorders present an opportunity to study early changes underlying neurocognitive impairment at both clinical and molecular levels, and to explore management strategies and future therapeutic opportunities.

COVID-19 and Toll-Like Receptor 4 (TLR4): SARS-CoV-2 May Bind and Activate TLR4 to Increase ACE2 Expression, Facilitating Entry and Causing Hyperinflammation

Mohamed M. Aboudounya et al.

Summary: Mortality in COVID-19 is caused by respiratory failure, heart failure, and sepsis/multiorgan failure. TLR4, an innate immune receptor, recognizes viral proteins and triggers immune responses. SARS-CoV-2 binds to TLR4, potentially leading to inflammation and multi-organ injury.

Summary: A substantial portion of COVID-19 patients develop long-lasting neuropsychiatric symptoms, potentially linked to chronic infection and inflammation of enterocytes in the small intestine. Co-expression analyses revealed that ACE2 in SARS-CoV2-infected enterocytes co-regulates with genes involved in dopamine metabolism and absorption of neurotransmitter precursors.

Targeting the coronavirus nucleocapsid protein through GSK-3 inhibition

Xiaolei Liu et al.

Summary: The nucleocapsid protein (N) of coronaviruses is essential for viral replication, and the phosphorylation of N by glycogen synthase kinase 3 (GSK-3) is crucial for its function. The study found that the N protein of SARS-CoV-2 may be sensitive to GSK-3 inhibitors like lithium, leading to reduced risk of COVID-19 in patients taking lithium. Inhibition of GSK-3 impairs viral transcription and replication, suggesting that targeting GSK-3 could be a potential approach to treat COVID-19 and future coronavirus outbreaks.

Summary: The study revealed that dopamine (DA) supply in peripheral organs originates from sympathetic neurons, with decreased TH protein and NA levels leading to a decrease in DA content in the heart and pancreas. TH-immunoreactive cells were found in the stomach and lung, showing a decreasing trend in TH protein levels but not in DA levels. Additionally, there was a significant correlation between DA content in the kidney and plasma DOPA concentration.
