Acidified Nitrite Contributes to the Antitumor Effect of Cold Atmospheric Plasma on Melanoma Cells

Tom Zimmermann et al.

Summary: CAP is a partially ionized gas with reported antitumor effects, but its active components and molecular mechanisms are not fully understood. This study characterized the impact of acidified nitrite on melanoma cells, demonstrating a synergistic effect with acidification leading to strong cytotoxicity. It also highlighted the importance of RNS during CAP treatment and evaluated important molecular mechanisms involved in the cancer cell response.

Tetraspanin 5 (TSPAN5), a Novel Gatekeeper of the Tumor Suppressor DLC1 and Myocardin-Related Transcription Factors (MRTFs), Controls HCC Growth and Senescence

Laura Schreyer et al.

Summary: Hepatocellular carcinoma (HCC) is a major cause of cancer-related deaths and understanding the molecular mechanisms underlying HCC growth and progression is crucial for developing better therapeutic options. Deleted in liver cancer 1 (DLC1) is a tumor suppressor gene in HCC, and our research identified TSPAN5 as a novel druggable target for HCC characterized by DLC1 loss, with potential implications for personalized medicine approaches in therapy.

Mechanisms of Cellular Senescence: Cell Cycle Arrest and Senescence Associated Secretory Phenotype

Ruchi Kumari et al.

Summary: Cellular senescence is a stable cell cycle arrest triggered in response to various stimuli, characterized by dynamic changes in senescent cells. It has both positive effects on tissue repair and anti-cancer mechanisms, but can also negatively impact organismal health. Multiple pathways, including p53/p21 and p16/pRB, play central roles in regulating this process.

Ouabain and chloroquine trigger senolysis of BRAF-V600E-induced senescent cells by targeting autophagy

Valentin L'Hote et al.

Summary: This study reveals that cardioglycosides such as ouabain can act as potent senolytics in BRAF-induced senescence by mediating signal transduction through Na,K-ATPase pump. Signal transduction is required for the senolytic effect of ouabain against BRAF-senescent cells.

Overcoming the senescence-associated secretory phenotype (SASP): a complex mechanism of resistance in the treatment of cancer

Cecilia R. Chambers et al.

Summary: Senescence is a cellular state where cells halt the cell cycle in response to stress, with senescent cancer cells and adjacent cells potentially remaining metabolically active and secreting pro-inflammatory factors known as the SASP. The SASP plays a critical role in tumorigenesis, affecting various processes and treatment efficacy. Future research on therapy-induced senescence and pharmacological manipulation may provide new avenues for cancer therapeutics.

Potential Use of Senolytics for Pharmacological Targeting of Precancerous Lesions

Tareq Saleh et al.

Summary: Senescence is a vital cell state that can be triggered by replicative exhaustion or oncogene hyperactivation, and senolytics are emerging drugs capable of selectively eliminating senescent cells. Evidence suggests that targeting oncogene-induced senescent cells with senolytics may provide a novel and less invasive approach to treat premalignant lesions and hinder their progression to cancer.

The Cancer SENESCopedia: A delineation of cancer cell senescence

Fleur Jochems et al.

Summary: The study comprehensively analyzed the transcriptome and senolytic responses in 13 cancer cell lines induced to undergo senescence, showing that the response to senolytic agents and the composition of the senescence-associated secretory phenotype in cancer cells are more influenced by the cell of origin than by the senescence trigger. Using machine learning, a gene expression classifier called SENCAN was established for detecting senescence in cancer cell samples, with the expression profiles and classifier accessible online as Cancer SENESCopedia.
