Neoadjuvant therapy versus direct to surgery for T4 colon cancer: meta-analysis

Flora Jung et al.

Summary: This review compared the oncological outcomes of neoadjuvant therapy and direct-to-surgery approaches in T4 colon cancer. The results showed that neoadjuvant therapy was associated with higher rates of margin-negative resection and improved 5-year overall survival compared to direct-to-surgery. There were no significant differences in cancer recurrence rates and postoperative morbidity between the two approaches.

Short-course radiotherapy followed by chemotherapy before total mesorectal excision (TME) versus preoperative chemoradiotherapy, TME, and optional adjuvant chemotherapy in locally advanced rectal cancer (RAPIDO): a randomised, open-label, phase 3 trial

Renu R. Bahadoer et al.

Summary: The RAPIDO trial aimed to reduce systemic relapses in locally advanced rectal cancer through short-course radiotherapy, chemotherapy, and delayed surgery. Results showed a decreased probability of disease-related treatment failure in the experimental group compared to standard care, with diarrhea being the most common adverse event and neurological toxicity being prominent in the standard care group during adjuvant chemotherapy.

Efficient detection and post-surgical monitoring of colon cancer with a multi-marker DNA methylation liquid biopsy

Shengnan Jin et al.

Summary: The mqMSP assay developed in this study showed high sensitivity and specificity in plasma samples from CRC patients, outperforming other methods in detecting early-stage and premalignant diseases, and predicting cancer recurrence. Additionally, the detection of ctDNA in preoperative plasma samples using this method can predict the likelihood of patient recurrence.

Preoperative detection of KRAS mutated circulating tumor DNA is an independent risk factor for recurrence in colorectal cancer

Yuki Nakamura et al.

Summary: This study investigated the relationship between preoperative KRAS mutated ctDNA status and postoperative recurrence in patients with CRC. The detection of KRAS mutation in ctDNA before surgery was found to be an independent factor associated with inferior recurrence-free interval and recurrence-free survival, suggesting its potential utility in guiding postoperative treatment decisions.

Minimal Residual Disease Detection using a Plasma-only Circulating Tumor DNA Assay in Patients with Colorectal Cancer

Aparna R. Parikh et al.

Summary: The study demonstrates that detection of persistent circulating tumor DNA in plasma shows favorable sensitivity and specificity for predicting recurrence after curative-intent surgery for colorectal cancer. Integrating analysis of genomic and epigenomic signatures enhances the accuracy of detection, supporting the potential clinical utility of plasma-only ctDNA minimal residual disease detection.

Summary: The detection of circulating tumor cells in patients with colorectal cancer before surgery is an independent and strong prognostic factor for overall survival. This study suggests that it could improve the identification of high-risk patients in future clinical trials.

Detection and characterization of lung cancer using cell-free DNA fragmentomes

Dimitrios Mathios et al.

Summary: Non-invasive assessment of cell-free DNA (cfDNA) provides an opportunity for cancer detection and intervention. Using a machine learning model and genome-wide analysis, the study successfully detected tumor-derived cfDNA in a prospective study of 365 individuals at risk for lung cancer. Combining clinical factors and CEA levels, the approach showed high accuracy in distinguishing lung cancer subtypes and the fragmentation score served as an independent prognostic indicator of survival.
