Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Optimized PD-L1 scoring of gastric cancer

Birgid Schoemig-Markiefka et al.

Summary: This study optimized PD-L1 assessment in gastric cancer using endoscopically available tissue and found that CPS values from four surface biopsies can accurately reflect the PD-L1 status of the entire tumor. Preoperative biopsies showed lower sensitivity compared to tissue microarrays (TMAs).

First-line nivolumab plus chemotherapy versus chemotherapy alone for advanced gastric, gastro-oesophageal junction, and oesophageal adenocarcinoma (CheckMate 649): a randomised, open-label, phase 3 trial

Yelena Y. Janjigian et al.

Summary: Nivolumab plus chemotherapy showed superior overall survival and progression-free survival compared to chemotherapy alone in previously untreated patients with advanced gastric, gastro-oesophageal junction, or oesophageal adenocarcinoma, indicating it as a new standard first-line treatment option with acceptable safety profile.

The newly identified MEK1 tyrosine phosphorylation target MACC1 is druggable by approved MEK1 inhibitors to restrict colorectal cancer metastasis

Dennis Kobelt et al.

Summary: The study identified MACC1 as a new substrate of MEK1, impacting cell motility, tumor growth, and metastasis. Targeting the MEK1/MACC1 axis provides new therapeutic options for cancer treatment, especially for high-risk patients for metastasis. This finding may have implications beyond colorectal cancer to other solid tumor entities.
