Type I interferon activates MHC class I-dressed CD11b+ conventional dendritic cells to promote protective anti-tumor CD8+ T cell immunity

Ellen Duong et al.

Summary: In this study, the researchers investigated the different roles of tumor-infiltrating dendritic cells (DCs) in anti-tumor immunity. By comparing spontaneously regressing and progressing tumors, they found that tumor-reactive CD8+ T cells were lost in progressing tumors but preserved in regressing tumors. They also identified a specific type of DCs (ISG(+) DCs) in regressing tumors that activated CD8+ T cells and played an important role in presenting tumor-derived antigens. Furthermore, they showed that exogenous IFN-b stimulation could restore the lost anti-tumor immunity in progressing tumors. These findings suggest that targeting this functional DC state may be a potential approach for the treatment of human diseases.

Summary: Allogeneic stem cell transplantation followed by DC vaccination shows potential for hemato-oncology patients, but obtaining sufficient numbers of natural DCs, particularly cDC1s, remains a challenge. A culture protocol using donor-derived HPCs has been developed to address this issue.

Novel Antigenic Targets of HPV Therapeutic Vaccines

Ditte Rahbaek Boilesen et al.

Summary: HPV infection is the leading cause of cervical and head and neck cancers globally, with vaccines and screening programs proving effective in prevention. However, cervical cancer remains a significant cause of morbidity and mortality, especially in developing countries. Therefore, the development of more effective therapeutic vaccines is crucial in combating HPV-related cancers.

Type 2 dendritic cells mediate control of cytotoxic T cell resistant tumors

Summary: DC2s, the majority of conventional DCs within most tumors, have the potential to initiate and sustain antitumor immunity, showing significant correlation with survival across multiple human cancers. Impairment of DC1s and enhancement of DC2 functionality leads to improved tumor control, highlighting the therapeutic and prognostic potential of DC2s within checkpoint blockade-resistant tumors.
