Summary: Evidence supports a role for estrogen related receptor-gamma (ESRRG) in the development of placental dysfunction in fetal growth restriction (FGR) and preeclampsia. ESRRG is highly expressed in normal placenta, reduced in FGR and preeclampsia, and its expression is altered by hypoxia. MiRNA or other potential upstream regulators may negatively influence ESRRG, contributing to placental dysfunction in FGR and preeclampsia, suggesting ESRRG pathway as a potential therapeutic target.

Prenatal Exposure to Retrorsine Induces Developmental Toxicity and Hepatotoxicity of Fetal Rats in a Sex-Dependent Manner: The Role of Pregnane X Receptor Activation

Summary: The study investigated the developmental and fetal hepatotoxicity induced by RTS, a type of PA, in pregnant Wistar rats. Results showed that prenatal RTS exposure caused reduced fetal bodyweights, decreased hepatocyte numbers, and potentiated hepatic apoptosis in fetuses, particularly females. RTS was found to be a PXR agonist in fetal liver and its toxicity was exacerbated in female fetuses due to increased CYP3A induction and self-metabolism.Activation or inhibition of AR affected RTS-mediated PXR activation, leading to differential susceptibility to RTS toxicity between male and female fetuses. These findings suggest that PXR may be a potential therapeutic target for PA toxicity.
