Cancer Statistics, 2021

Rebecca L. Siegel et al.

Summary: Every year, the American Cancer Society projects the numbers of new cancer cases and deaths in the United States, with the latest data showing a significant decline in lung cancer mortality, while prostate cancer mortality has plateaued and breast and colorectal cancer mortality have slowed. Improvements in treatment have accelerated progress against lung cancer, leading to a record drop in overall cancer mortality.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

A synergetic effect of BARD1 mutations on tumorigenesis

Wenjing Li et al.

Summary: Mutations in the BARD1 gene have been identified as risk factors in breast and ovarian cancers. Here the authors reveal a synergetic effect between the occurance of two cis mutations in the BARD1 gene affecting the cellular DNA damage response and consequently tumorigenesis.

Ovarian cancer population screening and mortality after long-term follow-up in the UK Collaborative Trial of Ovarian Cancer Screening (UKCTOCS): a randomised controlled trial

Usha Menon et al.

Summary: The long-term follow-up of the UK Collaborative Trial of Ovarian Cancer Screening (UKCTOCS) did not show a significant reduction in deaths due to ovarian or tubal cancer with screening. While there was an increase in early-stage cancer detection in the MMS group, it did not translate into lives saved, highlighting the importance of specifying cancer mortality as the primary outcome in screening trials. Consequently, general population screening for ovarian and tubal cancer cannot be recommended.

Clinical cancer genomic profiling

Debyani Chakravarty et al.

Summary: Profiling tumors by next-generation sequencing has the potential to improve diagnostic accuracy, assess heritable cancer risk, and guide treatment selection. Through integrative analyses of tumor and germline variants, as well as identifying mutational signatures that influence therapy response, the clinical utility of cancer genomic profiling is being enhanced. Rapid technological innovation and reduced sequencing costs have enabled the profiling of hundreds of cancer-associated genes as part of routine cancer care.

A single-cell and spatially resolved atlas of human breast cancers

Sunny Z. Wu et al.

Summary: This study presents a comprehensive transcriptional atlas of the cellular architecture of breast cancer, identifying recurrent neoplastic cell heterogeneity and new immune cell populations associated with clinical outcomes. A multi-omic atlas integrates single-cell RNA sequencing, spatial transcriptomics, and immunophenotyping to stratify breast cancer into nine ecotypes with unique cellular compositions and clinical outcomes.

Spatial omics and multiplexed imaging to explore cancer biology

Sabrina M. Lewis et al.

Summary: Understanding intratumoral heterogeneity through single-cell analyses has revealed new biomarkers, but lacks information on cellular location within the tumor microenvironment. New technologies enabling detection of cancer subclones in their native spatial context promise to drive the next generation of cancer research, diagnosis, and therapeutic strategies.

Integrating single-cell and spatial transcriptomics to elucidate intercellular tissue dynamics

Sophia K. Longo et al.

Summary: Integrating single-cell RNA sequencing with spatial transcriptomics allows for the localization of transcriptionally characterized single cells within their native tissue context. While scRNA-seq identifies cell subpopulations within tissue, it does not capture their spatial distribution or reveal local networks of intercellular communication. Recent techniques like multiplexed in situ hybridization and in situ sequencing, defined as high-plex RNA imaging, can help address these limitations. However, the need for approaches to integrate single-cell and spatial data remains, given that no current method provides as complete a scope of the transcriptome as scRNA-seq.

Targeting PUS7 suppresses tRNA pseudouridylation and glioblastoma tumorigenesis

Summary: TF in plasma serves as an independent prognostic marker for disease progression and relapse in HGS-EOC patients. After platinum-based treatment, ctDNA analysis shows changes in genomic amplification patterns, with serially collected ctDNA samples' TF outperforming CA-125 in predicting disease progression.
