A Unique Immune-Related Gene Signature Represents Advanced Liver Fibrosis and Reveals Potential Therapeutic Targets

Pil-Soo Sung et al.

Summary: This study aimed to identify a unique immune gene signature that distinguishes advanced liver fibrosis in chronic liver diseases and to reveal potential therapeutic targets. A five-gene signature (CHIT1_FCER1G_OSM_VEGFA_ZAP70) was identified that reliably differentiated low- and high-grade fibrosis. Among these genes, inflammation-associated genes showed lower expression in high-grade fibrosis, while genes associated with fibrogenic activity showed higher expression. Furthermore, meta-analysis revealed a potential druggable target.

Crosstalk between tumor-associated macrophages and neighboring cells in hepatocellular carcinoma

Pil Soo Sung

Summary: Tumor-associated macrophages (TAMs) play a crucial role in hepatocellular carcinoma (HCC) progression and treatment resistance by interacting with various cell populations in the tumor microenvironment.

Single-cell landscape of the ecosystem in early-relapse hepatocellular carcinoma

Yunfan Sun et al.

Summary: This study showed that early-relapse hepatocellular carcinoma (HCC) tumors exhibit innate-like CD8(+) T cells with overexpression of KLRB1 (CD161) and a low cytotoxic state, unlike the exhausted state seen in primary HCC. These cells were associated with a worse prognosis. Additionally, potential immune evasion mechanisms were identified in recurrent tumor cells, which dampen dendritic cell antigen presentation and recruit innate-like CD8(+) T cells.

Regulation of PD-L1 expression in the tumor microenvironment

Ming Yi et al.

Summary: PD-L1 expression is regulated by various factors, impacting the treatment effect and patient selection. Understanding the mechanisms of PD-L1 regulation helps enhance the efficacy of immunotherapy.

Integrative single-cell transcriptome analysis reveals a subpopulation of fibroblasts associated with favorable prognosis of liver cancer patients

Haiyang Wang et al.

Summary: Single-cell transcriptome analysis was used to investigate the differences in cellular and molecular components between liver tumors and normal livers, revealing an expansion of specific fibroblast populations in liver cancer. This study identified SPARCL1-positive fibroblasts as potentially originating from endothelial cells and impacting the survival of liver cancer patients. Characterizing the spatial distribution and clinical significance of these fibroblast subpopulations provides new insights into the alterations in the cell atlas of liver cancers.

Hepatic macrophages: Key players in the development and progression of liver fibrosis

Da Cheng et al.

Summary: Hepatic fibrosis is a common pathological process involving persistent liver injury and inflammatory responses, which can lead to serious liver diseases if left untreated. Hepatic macrophages play a crucial role in this process, contributing to liver inflammation, fibrosis progression, and fibrosis resolution.

NASH limits anti-tumour surveillance in immunotherapy-treated HCC

Dominik Pfister et al.

Summary: The study investigated the impact of NASH on immunotherapy response in HCC patients and found that NASH-HCC may be less responsive to immunotherapy. Aberrant T cell activation caused by NASH-related tissue damage led to impaired immune surveillance in HCC patients.

Cross-tissue single-cell landscape of human monocytes and macrophages in health and disease

Kevin Mulder et al.

Summary: This study integrated a large number of mononuclear phagocytes from 13 tissues to generate a MNP single-cell RNA compendium, revealing conserved gene signatures and specialized cell subsets across multiple tissues. Specific macrophage populations were found to be expanded in cancer and inflammation, with tumor-associated macrophage populations present in all neoplastic tissues.

Tim F. Greten et al.

Summary: The standard of care for advanced HCC has been transformed by the combination of immunotherapy and anti-VEGF therapy, showing improved overall survival rates. The use of immunotherapy in later stages of treatment has been proven safe and effective, with ongoing research focusing on novel combinations of ICI and TKI for earlier intervention in HCC treatment.

NASH limits anti-tumour surveillance in immunotherapy-treated HCC

Dominik Pfister et al.

Summary: The text discusses the impact of NASH on the development of HCC and the effect of immunotherapy, revealing the accumulation of exhausted and unconventionally activated CD8(+)PD1(+) T cells in NASH-affected livers. In preclinical models of NASH-induced HCC, anti-PD1 treatment led to an increase in HCC, with CD8(+) T cells contributing to this increase.
