Summary: Macaques are important animal models in human immunology and infectious disease research, but their small size poses challenges for studies requiring large numbers of cells. This report provides a detailed protocol for collecting HSCs and T-cells from cynomolgus macaques as small as 3 kg, allowing for more efficient and detailed analysis of these cells and their progeny blood cells. The use of the Terumo Spectra Optia apheresis system allows for the collection of high numbers of cells, facilitating research in transplantation, vaccinology, and HIV/AIDS.
