Protein profiling in unlocking the basis of varicocele-associated infertility

Effects of HIF-1α on Spermatogenesis of Varicocele Rats by Regulating VEGF/PI3K/Akt Signaling Pathway

Summary: In this study, CRISPR/Cas9 gene editing was used to silence the HIF-1α gene in varicocele (VC) rat testes, revealing the regulatory role of HIF-1α in spermatogenesis in V rats through the PI3K/Akt signaling pathway. The downstream gene VEGF was found to be regulated by HIF-1α under hypoxic conditions, influencing the apoptosis of spermatogenic cells.

This suggests melatonin as a potential treatment option for male infertility caused by heat-induced oxidative stress.

NLRP3 Inflammasome: A New Pharmacological Target for Reducing Testicular Damage Associated with Varicocele

Summary: In varicocele animals, lower testosterone levels, testes weight, NLRP3 inflammasome, IL-1 beta and caspase-1 increased gene expression were demonstrated, along with an increased number of apoptotic cells. PDRN alone significantly improved all considered parameters more than Se. The Se-PDRN association significantly improved all morphological parameters, significantly increased testosterone levels, and reduced NLRP3 inflammasome, caspase-1, and IL-1 beta expression, as well as TUNEL-positive cell numbers.

Assessment of MUSASHI 1 and MUSASHI 2 expression in spermatozoa and testicular tissue

Summary: This study assessed sperm MSI1 and MSI2, as well as sperm functional tests in infertile men with varicocele and fertile men. While varicocele was associated with lower sperm concentration and motility, and higher abnormal sperm morphology, lipid peroxidation, DNA fragmentation and protamine deficiency, the expression of MSI1 and MSI2 genes in sperm cells was not significantly affected. The findings suggest that varicocele does not impact the relative expression of these genes in spermatozoa, despite their important roles in spermatogenesis.

Summary: This article summarizes the impact of sperm DNA fragmentation in various clinical infertility scenarios and advances in sperm DNA fragmentation tests. Based on collected evidence, 41 recommendations were formulated, covering technical aspects, clinical thresholds, interpretation of results, indications for testing, and clinical management. The bulk of evidence supporting these recommendations has increased in recent years, although it is still of moderate to low quality.
