Clamping, bending, and twisting inter-domain motions in the misfold-recognizing portion of UDP-glucose: Glycoprotein glucosyltransferase

Carlos P. Modenutti et al.

Summary: This study presents crystal structures of Chaetomium thermophilum UGGT and molecular dynamics simulations showing conformational changes. The estimated Parodi limit of UGGT is around 70-80 angstroms based on the MD simulations, with relevance of this limit to UGGT activity in vivo suggested by distance distributions in glycoprotein crystal structures.

C-phycocyanin as a highly attractive model system in protein crystallography: unique crystallization properties and packing-diversity screening

Iosifina Sarrou et al.

Summary: The study reports and discusses the unique crystallization properties of the antenna protein C-phycocyanin (C-PC) from the thermophilic cyanobacterium Thermosynechococcus elongatus. C-PC crystallizes in a variety of conditions with diverse crystal dimensions and morphology, and the addition of small-molecule additives can reveal new crystal packings. This protein's high propensity to crystallize, combined with its natural blue color and fluorescence characteristics, make it an excellent candidate in crystallography for technical and methods development, as well as for understanding the fundamental principles of protein crystallography.

Structural basis of cytokine-mediated activation of ALK family receptors

Steven De Munck et al.

Summary: ALK and LTK are recently identified receptor tyrosine kinases involved in neural development, cancer, and autoimmune diseases, along with their activating cytokines ALKAL1 and ALKAL2. Structural studies reveal a unique architecture of the cytokine-binding segments, and propose a mechanistic blueprint for the complexes of ALK family receptors.

RING domains act as both substrate and enzyme in a catalytic arrangement to drive self-anchored ubiquitination

Leo Kiss et al.

Summary: Attachment of ubiquitin to proteins is a versatile and abundant posttranslational modification that affects outcomes in various physiological processes. Substrate binding can induce a specific RING topology that enables self-ubiquitination. Studies have provided insights into the chemical mechanism of ubiquitin conjugation and the formation of ubiquitin chains.

Data- and diversity-driven development of a Shotgun crystallization screen using the Protein Data Bank

Gabriel Abrahams et al.

Summary: Protein crystallization plays a critical role in macromolecular structure determination through X-ray diffraction. This study introduces a new screening method, Shotgun II, based on analysis of the Protein Data Bank, and shows the success of the previous Shotgun I approach. By applying a more effective algorithm for selecting conditions, the Shotgun method aims to cover the most diverse space of proteins found in the PDB.
