Loss of type I IFN responsiveness impairs natural killer cell antitumor activity in breast cancer

Damien J. Zanker et al.

Summary: This study underscores the importance of intact type I IFN signaling to NK cells in the regulation of tumorigenesis and metastasis, and suggests that the IFNAR status on NK cells should be considered when devising therapeutic strategies aimed at inducing systemic type I IFN signaling in breast cancer.

NK cell infiltration is associated with improved overall survival in solid cancers: A systematic review and meta-analysis

Sarah Nersesian et al.

Summary: NK cell infiltration in solid tumors is associated with decreased risk of death and can predict overall survival, with intraepithelial NK cell infiltration being a better prognostic indicator. Despite lower infiltration in advanced-stage and lower-grade tumors, NK cells still have prognostic benefits.

Prostate Cancer Peripheral Blood NK Cells Show Enhanced CD9, CD49a, CXCR4, CXCL8, MMP-9 Production and Secrete Monocyte-Recruiting and Polarizing Factors

Matteo Gallazzi et al.

Summary: This study characterized the phenotype and function of peripheral blood NK cells in prostate cancer patients, finding that they promote inflammatory angiogenesis in endothelial cells and modulate monocytes and macrophages towards an M2-like phenotype. These findings suggest a potential use of peripheral blood NK cell profiling in prostate cancer patients.

Multi-tissue single-cell analysis deconstructs the complex programs of mouse natural killer and type 1 innate lymphoid cells in tissues and circulation

Adelle P. McFarland et al.

Summary: NK cells and ILC1s are heterogenous innate lymphocytes characterized by specific gene expression programs in different tissues, including blood, spleen, and solid tumors. The programs of circulating NK cells were found to be reshaped in tumor-bearing mice, with conserved signatures similar to human NK cells. This study advances the understanding of the NK-ILC1 spectrum in both murine and human systems.

Summary: The study demonstrated that administering IL-15 as CIV-5 significantly expanded NK cells with increased cytotoxic functions. There were no dose-limiting toxicities in the CIV-5 regimen, and a substantial increase in CD8(+) T cells was observed.

Innate lymphoid cells and cancer: Role in tumor progression and inhibition

Xiaodong Yuan et al.

Summary: ILCs, including NK cells, ILC1s, ILC2s, ILC3s, and LTi cells, have been identified as important players in tumor progression and inhibition. While NK cells and ILC1s are primarily involved in antitumor activities, the roles of ILC2s and ILC3s in tumors are still unclear and influenced by various factors. Understanding the mechanisms that shape the phenotypes and responses of ILCs in the tumor microenvironment is crucial for potential cancer immunotherapy strategies.

Landscape of innate lymphoid cells in human head and neck cancer reveals divergent NK cell states in the tumor microenvironment

Uriel Y. Moreno-Nieves et al.

Summary: Despite the reported antitumor functions of NK cells, their contribution to tumor control in humans remains controversial due to incomplete understanding of the NK cell states within the tumor microenvironment (TME). This study demonstrates that peripheral circulating NK cells differentiate into two divergent pathways within the TME, resulting in different end states, with one having potent in vivo antitumor activity. This finding provides insight into the origin of certain NK cell states within the TME and identifies specific NK cell phenotypes with the greatest antitumor activity.

Combinatorial immunotherapy of N-803 (IL-15 superagonist) and dinutuximab with ex vivo expanded natural killer cells significantly enhances in vitro cytotoxicity against GD2(+) pediatric solid tumors and in vivo survival of xenografted immunodeficient NSG mice

Yaya Chu et al.

Summary: The study demonstrates that the combination of N-803 with dinutuximab and exPBNK cells significantly enhances the anti-tumor effects against GD2(+) solid tumors, by enhancing NK cell activity and anti-tumor actions, reducing the secretion of various factors from tumor cells, and extending the survival time of mice.

Summary: This study discusses the role of NK cells in angiogenesis disturbances associated with cancer and other angiogenesis-dependent diseases (ADDs). It explores the impact and transformation of NK cells in the TME and UME. Understanding these mechanisms can contribute to a better understanding of these diseases and may have therapeutic implications.
