Immune-related toxicities of checkpoint inhibitors: mechanisms and mitigation strategies

Ryan J. Sullivan et al.

Summary: The Review by Weber and Sullivan highlights the different types of immune-related adverse events associated with checkpoint inhibitors and discusses how their treatment and identification of possible predictive biomarkers may shed light on their mechanisms, as well as possible strategies and targets for prophylactic and therapeutic interventions to mitigate them.

Interferon-γ: teammate or opponent in the tumour microenvironment?

Angela M. Gocher et al.

Summary: IFN gamma plays pleiotropic roles in the tumor microenvironment, acting as both a 'teammate' to promote antitumor immune responses and an 'opponent' promoting tumor growth and suppressing immune responses.

Status of IL-4 and IL-10 driven markers in experimental models of Visceral Leishmaniasis

Srija Moulik et al.

Summary: The mRNA expression of IL-4- and IL-10-driven markers was enhanced in both experimental models of visceral leishmaniasis, demonstrating an increased expression of these markers in the presence of Leishmania donovani infection. However, only in the hamster model, a positive correlation between IL-10-driven markers and parasite load was observed in splenic tissues.

Cytokines in the generation and function of regulatory T cell subsets in leishmaniasis

Sanhita Ghosh et al.

Summary: CD4+ T regulatory cells (Tregs) are vital in maintaining self-tolerance and protecting host from inflammation-induced tissue damage. In Leishmania infection, Tregs' suppressive functions may exacerbate the disease and affect parasite clearance.

Unraveling the Role of Immune Checkpoints in Leishmaniasis

Rafael de Freitas e Silva et al.

Summary: Leishmaniasis is a major global health issue affecting millions of people annually, with immune checkpoints playing a key role in the infection process. While T cell receptors and their ligands have been studied, further research is needed to determine their exact role during Leishmania infections.

PD-1 Blockade Modulates Functional Activities of Exhausted-Like T Cell in Patients With Cutaneous Leishmaniasis

Renan Garcia de Moura et al.

Summary: Patients infected by Leishmania braziliensis exhibit increased expression of inhibitory checkpoint receptors (ICRs) like PD-1, PDL-1, PDL-2, TIM-3, and CTLA-4 in skin lesions. This study found a positive correlation between the proportions of PD1(+) T cells in circulation and in the lesions of the same patients. Additionally, while blocking PD-1 signaling in circulating T cells enhanced their function, the extent of PD-1 expression by circulating T cells did not correlate with the magnitude of skin lesions, suggesting exhausted-like T cells may not contribute to cutaneous immunopathology.

Leishmania donovani Metacyclic Promastigotes Impair Phagosome Properties in Inflammatory Monocytes

Christine Matte et al.

Summary: Leishmaniasis, caused by protozoan parasites, can have debilitating effects ranging from self-healing ulcers to life-threatening visceral pathologies. Research has shown that early-stage interactions between Leishmania and inflammatory monocytes can impact antimicrobial responses, with the parasites inhibiting microbicidal properties of phagosomes within the cells. This study found that Leishmania donovani metacyclic promastigotes can impair superoxide production and pH acidification within inflammatory monocytes, potentially contributing to the establishment of chronic disease.

Expression of Tim-3 drives phenotypic and functional changes in Treg cells in secondary lymphoid organs and the tumor microenvironment
