Summary: A new subgroup of T cells named peripheral helper T (Tph) cells has been implicated in autoimmune pathogenesis, with an imbalance of Tph cell subsets influencing the severity of immune-related diseases. Specifically, in psoriasis patients, Tph17 cells were found to be activated and correlated with disease severity, while plasma CXCL13 levels were elevated and associated with disease severity and Tph17 cell frequency. CD4(+)CXCR5(+) Tfh cells were also increased in psoriasis patients and positively correlated with disease severity, Tph17 cells frequency, and plasma CXCL13 levels, suggesting them as potential immunotherapeutic targets.

Summary: This study utilized single-cell RNA sequencing to compare the transcriptomic heterogeneity of CD8(+) T cells in psoriatic and healthy skin, identifying several inflammatory subsets enriched in psoriatic skin and two Tc17 cell subsets associated with disease severity. Despite high expression of coinhibitory receptors, the psoriatic Tc17 cells exhibited upregulated cytokine, cytolytic, and metabolic activity, distinguishing them from an exhaustion program observed in melanoma-infiltrating CD8(+) T cells.
