The 2021 WHO Classification of Tumors of the Thymus and Mediastinum: What Is New in Thymic Epithelial, Germ Cell, and Mesenchymal Tumors?

Alexander Marx et al.

Summary: This article provides an overview of the new developments in the fifth edition of the WHO classification of thymic epithelial tumors, mediastinal germ cell tumors, and mesenchymal neoplasms, focusing on diagnostic, molecular, and conceptual advances. Diagnostic advances include immunohistochemical characterization and the identification of genetic translocations. Molecular and tumor biological advancements include the finding that thymomas and most thymic carcinomas lack currently targetable mutations, but typically have a PD-L1(high) phenotype. Additionally, conceptual advancements for the classification of thymic NETs are discussed.

LncRNA and mRNA expression associated with myasthenia gravis in patients with thymoma

Ji Ke et al.

Summary: Overall, the study identified significant differential expression of 4360 lncRNAs and 2545 mRNAs in MG-positive thymoma. Overactivity of biological processes and molecular functions, such as dephosphorylation and hydrolysis, were found to potentially impact thymocyte survival during selection in the thymus.

Identification of differentially expressed circular RNAs associated with thymoma

Qingjun Wu et al.

Summary: This study investigated the expression profiles of circRNAs in thymoma patients and identified four significantly upregulated circRNAs that were positively correlated with immune imbalance in thymoma. Functional analysis revealed their involvement in cell-cell adhesion, MAPK pathways, and TNF pathway. Additionally, two parental genes of these circRNAs were found to be significantly related to the progression-free survival of thymoma patients.

LOXL1-AS1 promotes thymoma and thymic carcinoma progression by regulating miR-525-5p-HSPA9

Jin Wang et al.

Summary: The study showed that LOXL1-AS1 and HSPA9 are highly expressed in thymoma and thymic carcinoma, while miR-525-5p is expressed at low levels and associated with poor prognosis. LOXL1-AS1 acts as a sponge targeting miR-525-5p to promote HSPA9 expression, thereby promoting the growth and invasion of thymoma and thymic carcinoma cells.

METTL3-dependent MALAT1 delocalization drives c-Myc induction in thymic epithelial tumors

Alessia Iaiza et al.

Summary: This study revealed that METTL3 contributes to the phenotype of Thymic Epithelial Tumors, with overexpression in tumor tissue compared to normal tissue. Silencing of METTL3 in thymic carcinoma cells resulted in decreased cell proliferation and overall translation rate. Furthermore, it was found that METTL3 is responsible for the induction of c-MYC expression in TET cells, with c-MYC protein expression enabled by lncRNA MALAT1 methylated and delocalized by METTL3. Blocking c-MYC with JQ1 inhibitor in combination with METTL3 depletion showed cooperation in inhibiting proliferation and inducing cell death.
