Hsa_circ_0087352 promotes the inflammatory response of macrophages in abdominal aortic aneurysm by adsorbing hsa-miR-149-5p

Xiaoying Ma et al.

Summary: This study investigates the role of circRNA in abdominal aortic aneurysms (AAA) and macrophages. The researchers found that hsa_circ_0087352, a circRNA, is up-regulated in human AAA specimens and is related to the inflammatory response in THP-1 macrophages. Further experiments revealed that hsa_circ_0087352 promotes IL-6 transcription and the secretion of inflammatory cytokines by sponging endogenous hsa-miR-149-5p in macrophages, potentially through the ERK/NF-kappa B pathway. Additionally, hsa_circ_0087352 overexpression induces apoptosis in human vascular smooth muscle cells. These findings suggest the potential of hsa_circ_0087352 as a biomarker and therapeutic target for AAA.

Circular RNA PLCE1 promotes epithelial mesenchymal transformation, glycolysis in colorectal cancer and M2 polarization of tumor-associated macrophages

Bo Yi et al.

Summary: This study aimed to investigate the biological functions of circPLCE1 on epithelial mesenchymal transformation (EMT), glycolysis, and tumor-associated macrophage (TAM) polarization in colorectal cancer (CRC). The results showed that circPLCE1 expression was increased, while miR-485-5p expression was decreased in CRC. Knockdown of circPLCE1 inhibited CRC proliferation, glycolysis, and M2 macrophage polarization, while decreased miR-485-5p accelerated EMT, glycolysis, and TAM M2 polarization in CRC. Mechanism studies revealed that circPLCE1 acted as a competitive endogenous RNA to mediate ACTG1 expression through binding with miR-485-5p. In summary, circPLCE1 promotes EMT, glycolysis, and TAM M2 polarization through regulating the miR-485-5p/ACTG1 axis in CRC.

Identification and characterization of circRNAs encoded by MERS-CoV, SARS-CoV-1 and SARS-CoV-2

Zena Cai et al.

Summary: The life-threatening coronaviruses MERS-CoV, SARS-CoV-1 and SARS-CoV-2 have distinct abilities to encode circRNAs, with MERS-CoV exhibiting a much more prominent ability than SARS-CoV-1/2. Most viral circRNAs have low expression levels and are mainly expressed in the late stage of viral infection. Analysis suggests that viral circRNAs in MERS-CoV infections up-regulate genes related to mRNA splicing and processing in the early stage, and regulate genes involved in various functions in the late stage, while SARS-CoV-2 viral circRNAs down-regulate genes associated with metabolic processes and up-regulate genes related to cellular responses to oxidative stress in the late stage of infection.
