Broad proteomic screen reveals shared serum proteomic signature in patients with psoriatic arthritis and psoriasis without arthritis

Emmerik Leijten et al.

Summary: Novel serum proteins associated with the pathogenesis of PsA were identified by comparing PsA patients with healthy controls, psoriasis (Pso) patients, and ankylosing spondylitis (AS) patients. The study found 68 differentially expressed proteins in PsA compared to healthy controls, with the majority also dysregulated in Pso and/or AS. PsA and Pso patients shared a similar serum proteomic signature, suggesting a unified psoriatic disease spectrum.

Tissue-Resident Memory CD8+T Cells From Skin Differentiate Psoriatic Arthritis From Psoriasis

Emmerik F. Leijten et al.

Summary: Compared to psoriasis, patients with PsA have increased regulatory CD4+ T cells in peripheral blood and a specific population of CD8+CCR10+ T cells that are enriched in PsA. These CD8+CCR10+ T cells express a tissue-resident memory T cell transcriptional signature and a Tc2/22-like cytokine profile, suggesting a potential role in the development of PsA.

Drivers of Inflammation in Psoriatic Arthritis: the Old and the New

Summary: The production of IL-17 by both unconventional and conventional lymphocytes in psoriatic arthritis and ankylosing spondylitis may lead to new pathogenic pathways. These cells significantly contribute to IL-17-mediated pathology and have implications for potential therapeutic mechanisms.

Integrated analysis of multimodal single-cell data

Yuhan Hao et al.

Summary: The study introduces a weighted-nearest neighbor analysis framework to learn the relative utility of each data type in each cell, enabling integrative analysis of multiple modalities. Applied to a CITE-seq dataset, the method constructs a multimodal reference atlas of the circulating immune system and successfully identifies and validates previously unreported lymphoid subpopulations.

Early Origins of Psoriatic Arthritis: Clinical, Genetic and Molecular Biomarkers of Progression From Psoriasis to Psoriatic Arthritis

Stephen R. Pennington et al.

Summary: Although having skin/nail psoriasis is an important risk factor for developing psoriatic arthritis (PsA), it alone is not sufficient to identify which patients with psoriasis will develop PsA. Further molecular assessment of psoriasis patients may help refine the profile of those at risk of developing PsA, leading to the design of disease prevention studies and new treatment approaches.
