Drug-loaded polymeric nanoparticles: a review

Karine Cappuccio de Castro et al.

Summary: Polymeric nanoparticles are widely studied for drug encapsulation in modern medicine. The researchers focus on a formulation that can control drug administration at specific sites. Stimulus-responsive nanoparticles have attracted attention due to their long circulation time, ability to reach disease sites, and improved medication delivery to cells. This review summarizes the trends in drug delivery mechanisms and the impact of internal and external stimulus sources on stimulus-responsive nanocarriers. It also discusses the conventional and emerging methods for synthesizing polymeric nanoparticles.

In Vivo Antitumor and Antimetastatic Efficacy of a Polyacetal-Based Paclitaxel Conjugate for Prostate Cancer Therapy

Yolanda Fernandez et al.

Summary: The study demonstrates that tert-Ser-PTX conjugate can provide sustained release of PTX in a pH-responsive manner over 2 weeks, along with epimerization of PTX to 7-epi-PTX. Serum proteins can stabilize tert-Ser-PTX, showing enhanced stability in human serum compared to PBS. While in vitro efficacy assessments in PCa cells showed IC50 values higher than the free form of PTX, in vivo tolerability assays revealed that tert-Ser-PTX significantly reduces systemic toxicities associated with free PTX treatment.

Development, formulation, and cellular mechanism of a lipophilic copper chelator for the treatment of Wilson's disease

Laura Gauthier et al.

Summary: The study revealed that a new Cu(I) pro-chelator can protect hepatocytes against Cu-induced cell death and counteract cellular stresses induced by moderate copper concentrations. This pro-chelator proves to be efficient in delivering an active chelator intracellularly to cope with copper stress, surpassing current and under development chelators.

Design and optimisation of dendrimer-conjugated Bcl-2/xL inhibitor, AZD0466, with improved therapeutic index for cancer therapy

Claire M. Patterson et al.

Summary: The study presents the design and development of AZD0466, a drug-dendrimer conjugate, and uses preclinical and mathematical models to determine the optimal release rate of the drug for maximal therapeutic index. The optimized candidate shows improved efficacy and tolerability compared to AZD4320 alone, enabling progression of this promising dual Bcl-2/Bcl-x(L) inhibitor into clinical development.
