Human hepatic in vitro models reveal distinct anti-NASH potencies of PPAR agonists

Joost Boeckmans et al.

Summary: The study successfully reproduced key NASH characteristics in vitro and demonstrated differential effects of various PPAR agonists on combating NASH, with elafibranor showing the strongest anti-NASH effects. This further validates the importance of these in vitro models for preclinical investigation of anti-NASH compounds.

The Role and Mechanism of Oxidative Stress and Nuclear Receptors in the Development of NAFLD

Ting Hong et al.

Summary: The overproduction of ROS and oxidative stress play crucial roles in the pathogenesis of acute and chronic liver diseases. NAFLD is characterized as a redox-centered disease, and dysregulation of nuclear receptors (NRs) may be key in regulating this process. Targeting NRs could be a promising strategy for improving oxidation damage and treating NAFLD.

The acute transcriptional responses to dietary methionine restriction are triggered by inhibition of ternary complex formation and linked to Erk1/2, mTOR, and ATF4

Kirsten P. Stone et al.

Summary: Research has shown that dietary methionine restriction triggers an integrated stress response in the liver, which regulates gene expression through ATF4 and mTOR, Erk1/2 to adapt to reduced methionine intake, ultimately restoring hepatic methionine to normal levels.

Gallic Acid Ameliorated Impaired Lipid Homeostasis in a Mouse Model of High-Fat Diet-and Streptozotocin-Induced NAFLD and Diabetes through Improvement of β-oxidation and Ketogenesis

Jung Chao et al.

Summary: The study demonstrated the effects of gallic acid administration in a mouse model of obesity and hyperglycemia, showcasing its ability to alleviate high blood glucose levels and slow the progression of NAFLD. It also revealed that the hepatoprotective effect was partially due to its influence on metabolic pathways related to glucose, lipids, amino acids, purines, and pyrimidines, particularly through upregulation of beta-oxidation and ketogenesis.

Cystine and Methionine Deficiency Promotes Ferroptosis by Inducing B-Cell Translocation Gene 1

Il-Je Cho et al.

Summary: Cystine and methionine deficiency (CST/Met (-)) induces ferroptosis by activating ATF4-dependent BTG1 induction. This process depletes reduced and oxidized glutathione, leading to accumulation of reactive oxygen species and lipid peroxidation, ultimately causing necrotic cell death. Additionally, CST/Met (-) blocks global protein translation, increases expression of genes associated with the integrated stress response, and induces BTG1 expression.

Summary: Recent studies have shown promising effects of incretin-based therapies and PPAR agonists on NASH and T2D, highlighting the importance of targeting insulin resistance and metabolic dysfunction. However, the safety profile and limited impact on liver fibrosis of these drugs need to be further evaluated in long-term studies.

New drugs for NASH

Somaya A. M. Albhaisi et al.

Summary: NASH is a form of liver disease characterized by inflammation and hepatocyte injury in the presence of hepatic steatosis, potentially leading to cirrhosis. Although weight loss and lifestyle modifications are currently the first-line treatment, the development of effective drugs for NASH is urgently needed as these methods may not be effective in advanced cases. There has been significant growth in drug development targeting pathological pathways in NASH in the past decade.

Non-Alcoholic Steatohepatitis (NASH) - A Review of a Crowded Clinical Landscape, Driven by a Complex Disease

Julia M. Fraile et al.

Summary: Non-alcoholic steatohepatitis (NASH) is a common chronic liver condition with no FDA-approved therapies. Despite a growing market and clinical need, finding effective drugs has been challenging. Several drug candidates are in late-stage clinical trials, but the complex pathophysiology of NASH may require drug combination and targeting of different stages of the disease.
