Temozolomide: An Updated Overview of Resistance Mechanisms, Nanotechnology Advances and Clinical Applications

Raul Ortiz et al.

Summary: Temozolomide is an oral alkylating prodrug commonly used in the treatment of high-grade gliomas, with high oral bioavailability and the ability to cross the blood-brain barrier. However, despite its efficacy, patient survival remains relatively short, largely due to tumor drug resistance. Understanding the mechanisms of resistance is crucial for the development of new therapeutic strategies to improve clinical outcomes.

MSC-AS1 knockdown inhibits cell growth and temozolomide resistance by regulating miR-373-3p/CPEB4 axis in glioma through PI3K/Akt pathway

Chong Li et al.

Summary: In TMZ-resistant glioma, high expression of MSC-AS1 was found to be associated with reduced overall survival rate, while inhibition of MSC-AS1 decreased IC50 value of TMZ, promoted apoptosis, and affected PI3K/Akt pathway. Further investigation revealed that MSC-AS1 acted as a miR-373-3p sponge and miR-373-3p targeted CPEB4 directly, influencing TMZ sensitivity. Silencing miR-373-3p reversed the promoting effect of MSC-AS1 or CPEB4 knockdown on TMZ sensitivity, indicating a potential regulatory mechanism for chemoresistance.

Cell cycle arrest and apoptosis induction by Juniperus communis extract in esophageal squamous cell carcinoma through activation of p53-induced apoptosis pathway

Chia-Yu Li et al.

Summary: Juniperus communis extract has shown antitumor activity against esophageal squamous cell carcinoma (ESCC) cells by regulating the cell cycle and promoting apoptosis. It has higher selectivity for ESCC cells compared to clinical drug 5-fluorouracil (5-FU), and when combined with 5-FU, it synergistically inhibits ESCC cell proliferation. This suggests that JCo extract could be a potential natural therapeutic agent for esophageal cancer.

Cell-permeable transgelin-2 as a potent therapeutic for dendritic cell-based cancer immunotherapy

Hye-Ran Kim et al.

Summary: Transgelin-2 plays a crucial role in DC-based immunity, with deficiency leading to impaired DC functions in antigen presentation and T cell activation, ultimately resulting in the inability to suppress tumor growth and metastasis. Non-viral transduction of cell-permeable recombinant transgelin-2 enhances DC functions and shows potential clinical value in cancer immunotherapy.

PSMD12 promotes glioma progression by upregulating the expression of Nrf2

Zhongyong Wang et al.

Summary: PSMD12 is upregulated in glioma, especially in high-grade glioma, and is associated with poor prognosis in patients. Inhibition of PSMD12 expression can suppress the growth and invasion ability of glioma cells. PSMD12 promotes glioma development by increasing the expression of Nrf2.

Qiong-Ying Hu et al.

Summary: Studies have shown that malayoside has anti-NSCLC activity both in vitro and in vivo, mainly through activation of the MAPK-Nur77 signaling pathway, indicating that malayoside is a promising chemotherapeutic candidate for NSCLC.

TAGLN2 promotes the proliferation, invasion, migration and epithelial-mesenchymal transition of colorectal cancer cells by activating STAT3 signaling through ANXA2

Zhicheng Zhao et al.

Summary: The study revealed that TAGLN2 was highly expressed in CRC cells and its knockdown significantly inhibited proliferation, migration, invasion, and EMT of CRC cells while suppressing STAT3 phosphorylation. Overexpression of TAGLN2 promoted CRC progression by activating STAT3, and its effects were reversed by a STAT3 inhibitor. Additionally, ANXA2 was positively associated with STAT3 activation.
