A critical analysis of research methods and experimental models to study the root canal microbiome

Jose F. Siqueira Jr et al.

Summary: Endodontic microbiology focuses on the microbial causes and mechanisms of pulpal and periradicular inflammatory diseases. Research in this field has been ongoing for about 130 years, mainly aiming to establish the infectious causes of apical periodontitis and identify the microbial species associated with different types of endodontic infections. Various analytical methods have been used, each with its own strengths and limitations, to determine the efficacy of treatment procedures in eradicating or controlling infections. Suggestions are provided for improving study designs to address scientific questions more effectively and avoid compromising results, while also outlining potential areas for future research.

Present status and future directions: Microbiology of endodontic infections

Jose F. Siqueira Jr et al.

Summary: Apical periodontitis is an inflammatory disease caused by microbes, with bacteria being the most prevalent in endodontic infections. Bacterial infection in the root canal system is associated with necrotic pulp or previous treatment. Different types of apical periodontitis show varying microbial compositions, with bacterial interactions influencing the overall virulence of the community.

Adjunctive procedure with solvent mixtures in non-surgical endodontic retreatment: does it affect root dentin hardness?

Ines Ferreira et al.

Summary: This study compared the effects of different irrigation solvents and agitation techniques on dentin hardness, finding that certain irrigation protocols significantly decreased hardness while MEK/OOil increased it, with agitation not showing significant influence on hardness reduction.

Supplementary solvent irrigation efficacy on filling remnants removal comparing XP-endo Finisher R vs IrriSafe

Ines Ferreira et al.

Summary: This study compared the efficacy of XP-endo Finisher R and IrriSafe, with a solvent mixture of MEK/TCE, in removing root filling residues. The results showed that the final irrigation protocol with NaOCl and EDTA significantly reduced the volume of filling remnants, while the additional solvent mixture MEK/TCE increased the efficiency of reducing filling materials.

Summary: The study demonstrated that additional exposure of 24-hour preformed C. albicans biofilm to novel solvent mixtures MEK/TCE and MEK/OOil resulted in a significant reduction in biomass and cell viability, outperforming the conventional endodontic irrigating protocol using sodium hypochlorite and ethylenediaminetetraacetic acid.

The use of solvents for gutta-percha dissolution/removal during endodontic retreatments: A scoping review

Lara Dotto et al.

Summary: This scoping review aimed to explore the evidence on the use of solvents for gutta-percha dissolution during endodontic retreatments. Most studies suggested that solvents may complicate root canal cleanliness and facilitate gutta-percha remnants, recommending their avoidance except when necessary. High heterogeneity was observed, calling for further studies comparing different solvents in various clinical scenarios.
