Translocation of non-lytic antimicrobial peptides and bacteria penetrating peptides across the inner membrane of the bacterial envelope

Jakob Frimodt-Moller et al.

Summary: With the increase in multidrug-resistant pathogenic bacteria, the development of novel antimicrobials, including antimicrobial peptides and antimicrobial antisense agents, has become a focus of research. Non-lytic AMP and BPP-PMO/PNA conjugates are able to translocate across the cytoplasmic membrane via receptor-mediated transport, showing promising potential as new antibacterial agents.

Global burden of bacterial antimicrobial resistance in 2019: a systematic analysis

Christopher J. L. Murray et al.

Summary: Antimicrobial resistance (AMR) poses a significant threat to global human health, and this study provides the most comprehensive estimates of AMR burden to date. By estimating deaths and disability-adjusted life-years (DALYs) associated with bacterial AMR in 204 countries and territories in 2019, the study highlights the impact of resistance and the leading pathogen-drug combinations contributing to it.

Approaches for Systemic Delivery of Dystrophin Antisense Peptide Nucleic Acid in the mdx Mouse Model

Camilla Brolin et al.

Summary: The study compared the systemic administration of a 20-mer splice switching antisense PNA oligomer through intravenous and subcutaneous routes in mdx mice, showing slower excretion with subcutaneous administration and accumulation in kidneys and liver with depot technology release. Formulation of PNA as a duplex hybridization complex increased solubility, but none of the alternative administration methods resulted in significantly improved antisense activity. Further improvements in formulation or delivery methods may be necessary for enhanced pharmacokinetics and tissue targeting.

Uptake, Stability, and Activity of Antisense Anti-acpP PNA-Peptide Conjugates in Escherichia coli and the Role of SbmA

Niloofar Yavari et al.

Summary: The study suggests that PNA oligomers conjugated with bacteria penetrating peptides can inhibit bacterial growth by targeting essential genes. The stability and metabolism of the peptides should be considered in the design and analysis of antimicrobial peptides to ensure effective uptake and activity. Additionally, the intact peptide plays a crucial role in uptake but is not essential for intracellular antisense activity.

How to Enter a Bacterium: Bacterial Porins and the Permeation of Antibiotics

Jigneshkumar Dahyabhai Prajapati et al.

Summary: Traditional antibiotics are facing the challenge of Gram-negative bacteria developing resistance, prompting the urgent need for new antibacterial agents. Designing new antibiotic molecules that can rapidly penetrate Gram-negative bacteria and achieve lethal intracellular drug accumulation is a major challenge. The lack of simple experimental techniques to measure antibiotic uptake and local concentrations in subcellular compartments is a limiting factor.

Targeting of the Essential acpP, ftsZ, and rne Genes in Carbapenem-Resistant Acinetobacter baumannii by Antisense PNA Precision Antibacterials

Alireza Japoni Nejad et al.

Summary: In this study, a series of PNA-BPP conjugates targeting CRAB strains were designed and synthesized. These compounds demonstrated potent antimicrobial activity against CRAB, exhibiting complete growth inhibition at 1-2x MIC and sensitivity in 12 isolates. Additionally, the conjugates showed no bacterial membrane disruption activity and no cellular toxicity in HepG2 cells, indicating potential as precision designer antibiotics for specific treatment of CRAB infections.

Activating the Cpx response induces tolerance to antisense PNA delivered by an arginine-rich peptide in Escherichia coli

Jakob Frimodt-Moller et al.

Summary: Arginine-rich CPPs efficiently deliver antimicrobial peptide nucleic acid (PNA) to its intracellular mRNA target in bacteria, leading to a resistance mechanism that involves multiple genetic modifications, particularly activation of the Cpx-envelope stress response system.

Towards the sustainable discovery and development of new antibiotics

Marcus Miethke et al.

Summary: There is a growing demand for novel antimicrobials to treat infections caused by multidrug-resistant bacterial pathogens, but there is currently a lack of investment in this field. New agents with innovative chemistry and modes of action are urgently needed to combat the public health threat posed by antimicrobial resistance. The consortium proposes short-term and long-term solutions to overcome urgent limitations in research and funding, aiming to bridge the gap between academic, industrial, and political stakeholders to improve the ability to discover and develop new antibiotics.
