Prognosis and personalized treatment prediction in TP53-mutant hepatocellular carcinoma: an in silico strategy towards precision oncology

Summary: This study retrospectively analyzed 1135 HCC patients and developed a random forest-based prediction model to estimate TP53 mutational status, as well as a robust poor prognosis-associated signature which showed superior ability to predict survival in TP53-mutant patients. In silico screening revealed three targets and two agents that might have potential therapeutic implications in high-PPS patients, presenting a comprehensive view of potential treatment strategy.

Role of chromosome 1q copy number variation in hepatocellular carcinoma

Summary: This review summarizes literature reports of multiple genes proposed as possible drivers of 1q amplification in hepatocellular carcinoma, with TPM3 and NUF2 identified as candidates most highly associated with poorer outcomes. The most frequent location for gene amplification was found to be 1q22, consistent with the results of the literature search. Additional candidate genes involved in carcinogenesis were identified using publicly available copy number alteration data from The Cancer Genome Atlas project.

An artificial neural network model based on DNA damage response genes to predict outcomes of lower-grade glioma patients

Summary: This study identified aberrant expression of DNA damage response genes in LGG patients, which were associated with prognosis. An artificial neural network (ANN) model based on these genes was developed to predict patient outcomes, showing superior predictive ability compared to mutation markers. The model accurately identified patients with different prognoses and was associated with mutation status and immune microenvironment alterations. The ANN model has the potential to improve individualized therapies for LGG patients.

The identified subgroups showed different clinical characteristics and immune cell enrichment scores, with Group 4 having the poorest outcomes. Additionally, highly expressed genes in Group 4 were enriched in elements of the WNT signaling pathway.

Summary: This study divided HCC patients into DDR1 and DDR2 groups, showing that DDR2 patients had significantly prolonged OS when receiving ICIs, while DDR1 patients had better OS after traditional treatments. DDR status may serve as a potential biomarker to predict opposite clinical prognosis in immunotherapy and non-immunotherapy in HCC.
