A review of potential novel glaucoma therapeutic options independent of intraocular pressure

Wesam Shamseldin Shalaby et al.

Summary: Glaucoma is a progressive optic neuropathy that leads to retinal ganglion cell degeneration and visual field loss, making it the leading cause of irreversible blindness worldwide. Intraocular pressure (IOP) is the only modifiable risk factor known to slow or halt disease progression; however, glaucomatous damage persists in nearly 50% of patients despite significant IOP reduction. Various studies have explored non-IOP-related factors and interventions that can protect against or delay glaucomatous neurodegeneration and preserve vision. Experimental studies have shown effective neuroprotection in glaucoma, and clinical trials are ongoing to provide strong evidence of their effectiveness.

Understanding dexamethasone kinetics in the rabbit tear fluid: Drug release and clearance from solution, suspension and hydrogel formulations

Anusha Balla et al.

Summary: Controlling release and precorneal residence properties of topical formulations may improve ocular drug bioavailability and duration of action. In this study, dexamethasone was found to have prolonged exposure in tear fluid when administered as hydrogels and suspensions compared to a solution, indicating the potential for the design of dexamethasone delivery systems with improved ocular absorption and prolonged duration of action.

Drug Delivery Challenges and Current Progress in Nanocarrier-Based Ocular Therapeutic System

Md Habban Akhter et al.

Summary: Topical drug instillation is the preferred route for treating ocular ailments due to its noninvasive and convenient nature. However, the physiological barriers in the eye limit the drug availability. Innovative drug delivery systems, such as nanotechnology platforms, have been developed to overcome these barriers and improve drug bioavailability in ocular tissue. The rational design of ocular therapeutic systems is crucial for enhancing drug concentration and permeation in specific areas of the eye.
